The structures of compounds 3 were confirmed by X-ray crystallography. We propose a new protocol for synthesis of the arylidene-1H-imidazol-5(4H)-ones which is based onO-alkylation of amidoacetates using triethyloxonium tetrafluoroborate leading to carboximidates. These imidates have long been used in the synthesis of imidazolones by [2+3] cycloaddition reaction with Schiff bases. However, diversity of carboximidates prepared by the common techniques was limited. The protocol we proposed eliminated these limitations. Using this technique, a number of derivatives of the green fluorescent protein (GFP) and asFP chromophores were synthesized, and influence of the C2 residue on their optical properties was studied.

A series of substituted benzoate-bridged dichromium(II, II) complexes [Cr-2(RCO2)(4)(THF)(2)] ([Cr-2]), where RCO2- is substituted benzoate, was synthesized and its structural and magnetic properties were investigated. The orbital energies, as well as magnetic coupling energies, were also investigated using computational approaches. The HOMO/LUMO energy levels of the complexes are strongly dependent on the acidity, i.e., pK(a), of the corresponding benzoic acids (RCO2H), revealing a linear trend in the respective groups of non-ortho, mono-ortho, and bi-ortho substituted groups. The Cr center dot center dot center dot Cr magnetic coupling constant (E-S (T)) is little affected by pK(a); instead, the E-S (T) is associated with the HOMO/LUMO gap and strongly correlated with the Cr center dot center dot center dot Cr distance.

Benzotriazoles (4-6) were synthesized which were further reacted with different substituted benzoic acids and phenacyl bromides to synthesize benzotriazole derivatives (7-40). The synthetic compounds (7-40) were characterized via different spectroscopic techniques including EI-MS, HREI-MS, H-1-, and C-13 NMR. These molecules were examined for their anti-hyperglycemic potential hence were evaluated for alpha-glucosidase and alpha-amylase inhibitory activities. All benzotriazoles displayed moderate to good inhibitory activity in the range of IC50 values of 2.00-5.6 and 2.04-5.72 mu M against alpha-glucosidase and alpha-amylase enzymes, respectively. The synthetic compounds were divided into two categories A and B, in order to understand the structure-activity relationship. Compounds 25 (IC50 = 2.41 +/- 131 mu M), (IC50 = 2.5 +/- 1.21 mu M), 36 (IC50= 2.12 +/- 1.35 M), (IC50 = 2.21 +/- 1.08 mu M), and 37 (IC50 = 2.00 +/- 1.22 mu M), (IC50 = 2.04 +/- 1.4 mu M) with chloro substitution/s at aryl ring were found to be most active against alpha-glucosidase and alpha-amylase enzymes. Molecular docking studies on all compounds were performed which revealed that chloro substitutions are playing a pivotal role in the binding interactions. The enzyme inhibition mode was also studied and the kinetic studies revealed that the synthetic molecules have shown competitive mode of inhibition against alpha-amylase and non-competitive mode of inhibition against alpha-glucosidase enzyme. Eleven new tetrahydrobenzo[b]pyran derivatives were synthesized via a three component reaction of different aromatic aldehydes, methyl cyanoacetate and 1,3-cyclohexadione, with water as solvent under catalyst-free microwave irradiation. The structures of all the new molecules were well analysed and their structures established by using various spectral techniques (H-1 NMR, C-13 NMR, N-15 NMR and HRMS). Various advantages of reported protocol are the ease of preparation, short reaction times (10 min), aqueous solvent and excellent yields (89-98%). Additionally, this method provides a clean access to the desired products by simple workup.

A new metal-, oxidant-, and solvent-free ecofriendly domino method has been established for modular synthesis of a diverse range of medicinally promising hydroxyarylated unsymmetrical pyridines in good to high chemical yields with an excellent regioselectivity. This domino process involves a range of N-sulfonyl ketimines as C,N-binucleophiles, enolizable ketones, and aromatic/heteroaromatic aldehydes using ammonium acetate as an ideal promoter under neat conditions, which creates two new C-C bonds and one C-N bond. Notably, the neutral reaction conditions are mild enough to tolerate a range of functionalities and cover a variety of substrates, thus bestowing a powerful avenue to access tri- and tetrasubstituted pyridines including carbo- and heterocyclic fused ones. Interestingly, a practical, scalable, and high-yielding synthesis of pyridylphenol derivatives was successfully accomplished by our unique method.

A one-step synthesis of acyl phosphoramidates from a variety of functionalized acyl azides has been developed employing trimethylsilyl chloride as an activating agent in a modified Staudinger reaction. In the presented work, we report the synthesis of a series of 4-benzylidene-2-phenyl-5(4H)-imidazolone-based benzenesulfonamides 7a-f via the Erlenmeyer-Plochl reaction. All the prepared imidazolones 7a-f were evaluated as inhibitors of human (h) carbonic anhydrases (CA, EC 4.2.1.1) cytosolic isoforms hCA I and II, as well as transmembrane tumor-associated isoforms hCA IX and XII. All the tested hCA isoforms were inhibited by the prepared imidazolones 7a-f in variable degrees with the following KIs ranges: 673.2-8169 nM for hCA I, 61.2-592.1 nM for hCA II, 23-155.4 nM for hCA XI, and 21.8-179.6 nM for hCA XII. In particular, imidazolones 7a, 7e, and 7f exhibited good selectivity towards the tumor-associated isoforms (CAs IX and XII) over the off-target cytosolic (CAs I and II) with selectivity index (SI) in the range of 6.2-19.4 and 3.3-8, respectively. Moreover, imidazolones 7a-f were screened for their anticancer activity in one dose (10(-5) M) assay against a panel of 60 cancer cell lines according to US-NCI protocol. Furthermore, 7a, 7e and 7f were evaluated for their anti-proliferative activity against colorectal cancer HCT-116 and breast cancer MCF-7 cell lines. Furthermore, 7e and 7f were screened for cell cycle disturbance and apoptosis induction in HCT-116 cells. Finally, a molecular docking study was carried out to rationalize the obtained results.
