Downy mildew is a very important detrimental disease that lead reduced to fruits and vegetables. Due to the continuous growth of drug resistance, finding novel fungicides with dissimilar modes of function from present fungicides for controlling downy mildew are imminent. This work is an extension of our preceding research on the original triazole sulfonamide derivatives lead compound. Triazole sulfonamide as a remarkable nitrogen-containing heterocyclic compound opposed cucumber downy mildew (CDM) develops a quite vital part in the sphere of the study of new farm chemicals. The existing report designs a certain amount of 1,2,4-triazole-1,3-disulfonamide derivatives. Hydrazones have obtained extensive attention in the field of pharmaceutical due to its unique chemical structure and remarkable activity (insecticidal, antibacterial, antifungal and herbicidal). By means of coupling numerous hydrazone with triazole sulfonyl chloride groups, 24 novel derivatives were synthesized. Spectrum analysis of LC-MS, H-1 NMR and C-13 NMR were used for characterizing these new compounds. Compared with commercial Cyazofamid using bioassays, most of these compounds displayed preferable fungicidal activities. Moreover, compounds 8q illustrated the greatest CDM resistance (EC50 = 7.776 mg/L). Field efficacy trials revealed that compound 8q fungicidal activity was higher than the purchased agrochemical Cyazofamid and Amisulbrom. Thus, the research declared that 8q displayed a great potential for the application of fungicide against CDM.

Present work demonstrates synthesis and antibacterial property of new pyrazole derivatives. A series of new Curcumin based dihydropyrazoles has been synthesized with an objective to evaluate their antibacterial property. Dihydropyrazoles analogues were synthesized using Curcumin based chalcones and differently substituted phenylhydrazine derivatives. We used the previously designed Curcumin based chalconesto react with phenylhydrazine derivatives in ethanol in a catalyst free medium to afford new dihydropyrazole derivatives. Effect of substituent on reactivity was also studied. All the synthesized pyrazole analogues were characterized using proton and carbon NMR, Mass spectroscopy and IR techniques. Effect of substituent on reactivity was explained on the basis of electronic effect generated due to groups on phenyl ring. Presence of dd (double doublet) in proton NMR spectrum of Dihydropyrazoles was also explained due to presence of optically active carbon of pyrazole ring. The synthesized library was screened for their inhibitory activity against 4 different bacterial strains 1. E. Coli (ATCC 9637), 2. Pseudomonas aeruginosa (ATCC BAA-427), 3. Staphylococcus aureus (ATCC 25923) and 4. Klebsiella pneumonia (ATCC 27736). Out of all the compounds evaluated, the compounds that exhibited IC50 value greater than 50 mu M, were considered to be inactive. We established an important SAR based on the structure dependent inhibitory potential of screened dihydropyrazoles. Two compounds 4e and 4t having nitro and benzyl substitution respectively were showing the best inhibitory potential against Gram Positive bacterial strain Staphylococcus aureus with MIC value of 1.56 mu g/ml. Compounds having Chloro and Methoxy substitution were found to be less effective against screened bacterial strains. Compounds 4a and 4b were selective towards Staphylococcus aureus species with the MIC value of 1.56 mu g/ml for each. These pyrazole analogues were not showing inhibitory potential against other screened bacterial strains.

Scaffolds containing one or more heteroatoms are ubiquitous in nature and are responsible for almost every biological processes in flora and fauna. The fact that heterocyclic cores have high propensity for biological targets, several nitrogen, oxygen, and sulfur-containing heterocyclic compounds have been reported in the last few decades. One such intriguing class is 4-thiazolidinone that exhibit diverse range of pharmacological activities. In the present study, we report synthesis, characterization, molecular docking, and anticancer evaluation of 10 new 4-thiazolidinone analogues (5-14). One of the compounds (11) was structurally characterized using single crystal X structure. Anticancer evaluation against hepatocellular carcinoma cell line (HepG2) revealed that compound (7) was the most potent (IC50=75 mu M) while other showed moderate to low activity (85-530 mu M). To underpin the druggability and possible modes of action, drug-likeness was determined and docking studies were carried out against beta-carbonic anhydrase receptor (PDB ID: 3IA). Attempts have also been made to establish a structure-activity relationship of the reported compounds.

Benzotriazoles (4-6) were synthesized which were further reacted with different substituted benzoic acids and phenacyl bromides to synthesize benzotriazole derivatives (7-40). The synthetic compounds (7-40) were characterized via different spectroscopic techniques including EI-MS, HREI-MS, H-1-, and C-13 NMR. These molecules were examined for their anti-hyperglycemic potential hence were evaluated for alpha-glucosidase and alpha-amylase inhibitory activities. All benzotriazoles displayed moderate to good inhibitory activity in the range of IC50 values of 2.00-5.6 and 2.04-5.72 mu M against alpha-glucosidase and alpha-amylase enzymes, respectively. The synthetic compounds were divided into two categories A and B, in order to understand the structure-activity relationship. Compounds 25 (IC50 = 2.41 +/- 131 mu M), (IC50 = 2.5 +/- 1.21 mu M), 36 (IC50= 2.12 +/- 1.35 M), (IC50 = 2.21 +/- 1.08 mu M), and 37 (IC50 = 2.00 +/- 1.22 mu M), (IC50 = 2.04 +/- 1.4 mu M) with chloro substitution/s at aryl ring were found to be most active against alpha-glucosidase and alpha-amylase enzymes. Molecular docking studies on all compounds were performed which revealed that chloro substitutions are playing a pivotal role in the binding interactions. The enzyme inhibition mode was also studied and the kinetic studies revealed that the synthetic molecules have shown competitive mode of inhibition against alpha-amylase and non-competitive mode of inhibition against alpha-glucosidase enzyme. A new group of potential pFOX inhibitors among linear methoxyphenyltriazaalkanes were designed and synthesized. Cardiotropic activity in rodent experiments was found for most of the synthesized compounds. The most active compound N-1-(2,3,4-trimethoxybenzyl)-N-2-{2-[(2,3,4-trimethoxybenzyl)amino]ethyl}-1,2-ethanediamine trihydrochloride (LM-1) combined anti-ischemic, antiarrhythmic, and antifibrillatory activities (1 mg/kg, i.v.) and low toxicity (LD50 = 119 mg/kg, mice, i.p.) and was selected for development as a potential cardiotropic drug.

Unprecedented triazinyl-isoxazoles were afforded via an effective cycloaddition reaction between nitrile oxides and the scarcely described 2-ethynyl-4,6-dimethoxy-1,3,5-triazine as dipolarophile. The biological evaluation of the newly synthesized compounds showed that the inhibition of human farnesyltransferase by zinc complexation could be improved with triazine-isoxazole moieties. The replacement of the isoxazole unit by a pyrrolidin-2-one was detrimental to the inhibitory activity while the pyrrolidin-2-thione derivatives conserved the biological potential. Predicting the acute toxicity of a large dataset of diverse chemicals against fathead minnows (Pimephales promelas) is challenging. In this paper, 963 organic compounds with acute toxicity towards fathead minnows were split into a training set (482 compounds) and a test set (481 compounds) with an approximate ratio of 1 : 1. Only six molecular descriptors were used to establish the quantitative structure-activity/toxicity relationship (QSAR/QSTR) model for 96 hourpLC(50)through a support vector machine (SVM) along with genetic algorithm. The optimal SVM model (R-2= 0.756) was verified using both internal (leave-one-out cross-validation) and external validations. The validation results (q(int)(2)= 0.699 andq(ext)(2)= 0.744) were satisfactory in predicting acute toxicity in fathead minnows compared with other models reported in the literature, although our SVM model has only six molecular descriptors and a large data set for the test set consisting of 481 compounds.

A series of tetrahydro-ss-carbolines substituted with an alkyl or acyl side chain was synthesized and screened for its antifungal activity against plant pathogenic fungi (Bipolaris oryzae, Curvularia lunata, Fusarium semitectum, and Fusarium fujikuroi). The structure activity relationship revealed that the substituent at the piperidine nitrogen plays an important role for increasing antifungal activities. In this series, 2-octyl-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole (3g) displayed potent antifungal activities with a minimum inhibitory concentration of 0.1 mu g/mL, including good inhibitory activity to the radial growth of fungus at a concentration of 100 mu g/mL compared to amphotericin B.
