Herein, we present the first report on TiO2 NPs based hybrid nano-reactor designed and synthesized via N-(2-amino ethyl)-3-amino propyl trimethoxy silane (AAPTMS) functionalization of graphene oxide (GO). The assynthesized nano-reactor was characterized by means of several analytical-cum-spectroscopic techniques that support the successful attempt for the synthesis of TiO2@AAPTMS-rGO. Further, the present catalyst was evaluated for its catalytic performance towards an expedient access to biologically relevant synthesis of 2,4,5-triaryl imidazoles. The functional sites of TiO2@AAPTMS-rGO direct the catalysis from the reactant to product with high atom economy, shorter reaction time and excellent yields. Moreover, the present catalyst TiO2@AAPTMS-rGO shows promising reusability for several catalytic cycles. It also provides a robust approach for 2,4,5-triaryl imidazoles with improved performance and without significant loss in catalytic activity. Further, this catalyst can be upgraded for other organic transformations leading to numerous pharmaceuticals and environment benefits.

A thermodynamically controlled and reversible nucleophilic addition of a monofluorinated sulfone, alpha-fluoro-alpha-phenylthio-alpha-phenylsulfonylmethane (FTSM), to aldehydes has been developed, which allows the efficient synthesis of beta-fluorinated carbinols with high diastereoselectivity. Early studies demonstrated that over expression of indoleamine 2,3-dioxygenase (IDO1) in tumor microenvironment results in tumor immune escape. Herein, in order to simplify the structure of two kinds of IDO1 inhibitors from marine alkaloid, Exiguamine A and Tsitsikammamines, we designed, synthesized a series of 1H-indole-4,7-dione derivatives and evaluated their inhibitory activity in IDO1 enzyme and in IFN-gamma stimulated Hela cells in vitro. The structure-activity relationship demonstrated that 5-(pyridin-3-yl)-1H-indole-4,7-dione is a promising scaffold for IDO1 inhibitors and most compounds with this core showed moderate inhibition potency at micromole level. Our further enzyme kinetics experiments reveal that these new developed compounds might act as reversible competitive inhibitors of IDO1.

Four series of berberine derivatives were designed and synthesized. All the synthetic compounds were screened for in vitro glucose consumption activity in HepG2 cell lines. The results showed that most of the tested compounds exhibited potent hypoglycemic activity, and the most potent compound 20b exhibited its potency by 3.23-fold of berberine, 1.39-fold of metformin and 1.20-fold of rosiglitazone, respectively. Western blot assay indicated these novel berberine-based derivatives executed their glucose-decreasing activity via the activation of AMPK pathway.

Up to 45 % of deaths in developed nations can be attributed to chronic fibroproliferative diseases, highlighting the need for effective therapies. The RGD (Arg-Gly-Asp) integrin alpha v beta 1 was recently investigated for its role in fibrotic disease, and thus warrants therapeutic targeting. Herein we describe the identification of non-RGD hit small-molecule alpha v beta 1 inhibitors. We show that alpha v beta 1 activity is embedded in a range of published alpha 4 beta 1 (VLA-4) ligands; we also demonstrate how a non-RGD integrin inhibitor (of alpha 4 beta 1 in this case) was converted into a potent non-zwitterionic RGD integrin inhibitor (of alpha v beta 1 in this case). This work reports the synthesis of a novel quinolin-4(3H)-one based Schiff base ligand 3-[(E)-(2,5-dimethox-yphenyl)methylidene]amino-2-methylquinazolin-4(3H)-one (DMPAQ) and its coordination complexes of the type [M(DMPAQ)(phen)]X (1a-1c), where M = Cu(II), Zn(II) and Cd(II) ions, respectively, phen = 1,10-phe-nanthroline. All the synthesized compounds were characterized using UV-Visible, elemental analysis, FT-IR, H-1 NMR, Mass spectroscopy and TGA techniques. The triclinic structure of the DMPAQ is determined by employing single crystal X-ray crystallographic analysis. The characterization results suggested that the ligand, DMPAQ is bidentate and coordinate to the metal center through the lactum oxygen and the azomethine nitrogen. The synthesized DMPAQ ligand and complexes (1a-1c) were screened for their in vitro anticancer activity against the human breast adenocarcinoma cell line, MCF-7. The complexes 1a and 1b displayed significant anticancer ac-tivity against MCF-7 cells even at lower GI50 value (GI(50) = 0.016 mu M) than the standard drug doxorubicin (GI(50) = 0.018 mu M). Further, we have performed computational DFT studies on the chemical reactivity of the ligand and the three complexes by means of Conceptual Density Functional Theory (CDFT) through the Koopmans in DFT (KID) approximation to support the experimentally obtained results.

Totally colorless, transparent, and highly refractive poly(phenylene thioether ether)s (PPTEs) containing thianthrene-2,7-disulfanyl moiety were developed in this study. A new 4,4′-diol aromatic compound, 2,7-(4,4′-diphenol)thiothianthrene (DPTT), with a high sulfur content was designed and synthesized to develop polymers with high refractive indices and high thermal stabilities. The PPTEs were prepared by polycondensation of DPTT with organic dihalogen compounds, such as 2,6-difluorobenzonitrile (DFBN) and 4,6-dichloro-2-(methylthio) pyrimidine (DCMP). The obtained polymers showed good thermal stabilities such as relatively high glass transition temperatures (T(g)s) in the range of 153-176 degrees C and 5% weight loss temperatures (T-5%) over 344 degrees C. The cut-off wavelengths (lambda(0)) of the PPTE films are shorter than 343 nm, and they have a transmittance higher than 93% at 550 nm. The combination of the pyrimidine and high sulfur-containing thianthrene derivatives provides the PPTEs with a high refractive index of 1.7204 and a small birefringence of 0.0106 at 637 nm.

This report details a decarboxylative cross-coupling of (hetero)aryl carboxylates with iodoarenes in the presence of a gold catalyst (>25 examples, up to 96% yield). This reaction is site specific, which overcomes prior limitations associated with gold catalyzed oxidative coupling reactions. The reactivity of the (hetero)aryl carboxylate correlates qualitatively to the field effect parameter (F-ortho). Reactions with isolated gold complexes and DFT calculations support a mechanism proceeding through oxidative addition at a gold(I) cation with decarboxylation being viable at either a gold(I) or a silver(I) species.
