A diverse range of ureas, N-acylureas, carbamates, and thiocarbamates has been synthesized in good to excellent yields by reacting N-acylbenzotriazoles individually with amines or amides or phenols or thiols in the presence of diphenylphosphoryl azide (DPPA) as a suitable azide donor in anhydrous toluene at 110 degrees C for 3-4 hours. In this route, DPPA was found to be a good alternative to trimethylsilyl azide and sodium azide for the azide donor in Curtius degradation. The high reaction yields, one-pot and metal-free conditions, straightforward nature, easy handling, use of readily available reagents, and in many cases avoidance of column chromatography are the notable features of the devised protocol.

In this paper, solar binary chemical depolymerization, that is Solar Thermal Electrochemical Process (STEP), was implemented for an effective breaking of lignin into small molecules and hydrogen. Compared with the conventional unitary chemical thermolysis, solar binary chemical depolymerization of lignin has high efficiencies of the liquefaction and gasification with the low coke, and accompanied by the abundant production of hydrogen. And the reaction temperature of the STEP process was greatly lowered by an intervention of the electrolysis. The results showed that the total conversion and liquefaction of the lignin yielded 87.22% and 57.72% under a constant current of 0.4 A at 340 degrees C. Further characterizations show that lignin has been successfully decomposed into small molecules with high added-value and hydrogen by a combination of the thermolysis and electrolysis. And the particle size of aggregates and the color degree in the lignin aqueous solution was obviously decreased after the STEP process.

Aurora and polo-like kinases control the G2/M phase in cell mitosis, which are both considered as crucial targets for cancer cell proliferations. Here, naphthalene-based Aurora/PLK coinhibitors as leading compounds were designed through in silico approach, and a total of 36 derivatives were synthesized. One candidate (AAPK-25) was selected under in vitro cell based high throughput screening with an IC50 value = 0.4 mu M to human colon cancer cell HCT-116. A kinome scan assay showed that AAPK-25 was remarkably selective to both Aurora and PLK families. The relevant genome pathways were also depicted by microarray based gene expression analysis. Furthermore, validated from a set of in vitro and in vivo studies, AAPK-25 significantly inhibited the development of the colon cancer growth and prolonged the median survival time at the end of the administration (p < 0.05). A series of benzophenone derivatives bearing naphthalene moiety were designed, synthesized, characterized by H-1 NMR, C-13 NMR, and HRMS and evaluated for their antiproliferative activity against human breast cancer cell line (MCF-7). Most of the tested derivatives showed good to moderate cytotoxicity against MCF-7 cell line. Among them, compound 4u (IC50 = 1.47 +/- 0.14 mu M) was found to be the most active compound, which is more active than the standard drug cisplatin (IC50 = 15.24 +/- 1.27 mu M). In vitro tubulin polymerization in-hibition assay, EBI competition assay, cell cycle analysis, and cell apoptosis assay identified that compound 4u was a new tubulin polymerization inhibitor by targeting the colchicine binding site. Besides, molecular docking study showed that compound 4u has high binding affinities with the colchicine binding site of tubulin through hydrogen bond, cation-pi, and hydrophobic interaction.

In this work three novel series of c-Met/HDAC bifunctional inhibitors were designed and synthesized by merging pharmacophores of c-Met and HDAC inhibitors. The most potent compound 11j inhibited c-Met kinase and HDAC1 with IC50 values of 21.44 and 45.22 nM, respectively. In addition, 11j showed efficient antiproliferative activities against both MCF-7 and A549 cells with greater potency than the reference drug SAHA and Cabozantinib. This work may lay the foundation for developing novel dual c-Met/HDAC inhibitors as potential anticancer therapeutics.

Experimental solubilities have been determined by spectrophotometric methods for 1-chloro-4-bromobenzene, 1,4-dibromobenzene, 1,4-dichloro-2-nitrobenzene, benzoic acid, 4-nitrobenzoic acid, 3,5-dinitrobenzoic acid, 2-methoxybenzoic acid, 4-methoxybenzoic acid, 3-chlorobenzoic acid, 3,5-dinitro-2-methylbenzoic acid, 2-methylbenzoic acid, 3-methylbenzoic acid, 3,4-dichlorobenzoic acid, benzoin, 4-tert-butylbenzoic acid, 4-chlorobenzoic acid, 3,4-dimethoxybenzoic acid, 3,4,5-trimethoxybenzoic acid and o-acetoacetaniside dissolved in 2-ethyl-1-hexanol at 298.15 K. Our experimental measurements, combined with published solubility data retrieved from the published chemical and engineering literature, were used to derive Abraham model correlations for solute transfer into 2-ethyl-1-hexanol from both water and from the gas phase. The derived Abraham model correlations were found to back-calculate the measured experimental values to within +/- 0.12 log units (or less).

An approach to the generation of ultra-large chemical libraries of readily accessible (REAL”) compounds is described. The strategy is based on the use of two- or three-step three-component reaction sequences and available starting materials with pre-validated chemical reactivity. After the preliminary parallel experiments, the methods with at least similar to 80% synthesis success rate (such as acylation – deprotection – acylation of monoprotected diamines or amide formation – click reaction with functionalized azides) can be selected and used to generate the target chemical space. It is shown that by using only on the two aforementioned reaction sequences, a nearly 29-billion compound library is easily obtained. According to the predicted physico-chemical descriptor values, the generated chemical space contains large fractions of both drug-like and beyond ruleof-five members, whereas the strictest lead-likeness criteria (the so-called Churcher’s rules) are met by the lesser part, which still exceeds 22 million.

Text. A general method for the synthesis of structural diversity and complexity of azepines from aldimine esters and beta ‘-acetoxy allenoates is reported. Under phosphine catalysis, a [4+3] cycloaddition for the formation of 1,3-dihydro-2H-azepine-2,2,4-tricarboxylates was achieved with broad substrate scope under mild reactions. A switchable process was given and a variety of important 2,3-dihydrochromeno[4,3-b]azepin-6(1H)-ones were selectively formed when the reaction was performed in the presence of Cs2CO3 and PPh3, which involved an intramolecular ester group migration and subsequent lactonization of 1,3-dihydro-2H-azepine-2,2,4-tricarboxylates. Besides easy handle process, high synthetic value of resulting products, it is worth to note that this work showed the novel example of 1,5-ethoxycarbonyl migration.
