The catalytic efficiency of zeolitic imidazolate framework (ZIF-8) has been explored by making it multifunctional. Dual active sites were incorporated such as acid (dodecatungstophosphoric acid, DTP) and metal (Pd degrees) to prepare 5% Pd-DTP@ZIF-8. DTP was encapsulated inside the cage of ZIF-8 by in-situ and bottle around the ship approach whereas Pd was loaded ex-situ by simple wet impregnation method. The catalytic efficiency was tested for one pot synthesis of 3-phenyl propyl benzoate (3-PPB), a perfumery compound, from cinnamyl alcohol and benzoic anhydride. Trans-esterification of cinnamyl alcohol with benzoic anhydride gives cinnamyl benzoate which on further hydrogenation gives 3-PPB. Three different supports were screened such as ZIF-8, K10 and MCF out of which ZIF-8 showed the maximum activity because of its high surface area and smaller pore diameter. Further Pd, Ni and Cu metals were studied for selective hydrogenation of C=C bond among which 5% Pd-DTP@ZIF-8 gave almost 98% conversion of cinnamyl benzoate to 3-PPB with 93% selectivity. Fresh and spent catalysts were characterized by various techniques. 5% Pd-DTP@ZIF-8 showed anti-leaching property with great thermal stability. The turn over frequency and turn over number of the catalyst was observed to be 0.058 s(-1) and 835, respectively. A kinetic model was developed with good fit using LHHW mechanism and the activation energy calculated as 17.45 kcal/mol for hydrogenation step. Thus, the reaction was found to be kinetically controlled. The entire process is green.

A collection of N-substituted quinolin-2(1H)-ones were screened against a panel of clinically relevant protozoa (Leishmania, Trypanosoma and Acanthamoeba). Three quinolin-2(1H)-one compounds were identified as selective anti-Acanthamoeba agents. Further assessment revealed that these compounds were active against both trophozoite and cyst forms of A. castellanii Neff, and caused protozoa death via apoptosis. The data presented herein identify N-acyl quinolin-2(1H)-ones as a promising new class of selective anti-Acanthamoeba agents.

Competitive reactions, on the surface of plasmonic nanostructures, allow exploiting SERS signals for quantitative Therapeutic Drug Monitoring. As an example, the concentration of Erlotinib, an anti-EGFR small molecule, used for the treatment of non-small cell lung and pancreatic cancer, is determined. The numerous side effects and the variability of patient responses make Erlotinib a good candidate for monitoring. The new SERS based sensor can estimate Erlotinib down to nanomolar concentration and is based on the chemical reaction of the drug and of a competitor SERS reporter on the surface of gold nanostructures. Colloid solutions of naked gold nanoparticles obtained by laser ablation in solution were used for obtaining nanostructures with very efficient hot spots for SERS and with a clean surface for chemistry. Transition metal-catalyzed cross-couplings have been widely employed in the synthesis of many important molecules in synthetic chemistry for the construction of diverse C-C bonds. Conventional cross-coupling reactions require active electrophilic coupling partners, such as organohalides or sulfonates, which are not environmentally friendly enough. Herein, we disclose the first nickel-catalyzed Suzuki-Miyaura cross-coupling of aryl anhydrides and arylboronic acids for the synthesis of biaryls in a decarbonylation manner. The reaction tolerates a wide range of electron-withdrawing, electron-neutral, and electron-donating substituents in this process.

Highly enantioselective desymmetrisation of Fittig’s lactones with alcohols is promoted by bifunctional cinchona squaramides. The reactions were carried out with monodeuterated methanol to detect possible hidden racemisation of the stereogenic centre. Current evidence suggests that racemisation was not a relevant process for most substrates; partial erosion of enantioselectivity was only detected with ortho-substituted aryl derivates. The resultant glutaric acid derivatives possess a scaffold that is common in natural products and the compounds are also useful chiral building blocks for further synthetic endeavours.

A highly enantioselective alpha-ketol rearrangement has been developed. In the presence of a chiral Cu-bisoxazoline complex, achiral beta-hydroxy-alpha-dicarbonyls were isomerized to chiral alpha-hydroxy-beta-dicarbonyls and their bicyclic derivatives in excellent yields and enantioselectivities. Enantioenriched 2-acyl-2-hydroxy cyclohexan-1-ones, dihydroxyhexahydrobenzofuranones, and dihydroxyhexahydro-cycloheptafuranones, with up to three stereocenters, were readily prepared from achiral starting materials in one operation. The reaction is applicable to the desymmetrization of meso substrates and kinetic resolution of racemic alcohols.

Inorganic persulfate salts were identified as efficient reagents for the oxidative aromatization of 3,4-dihydroquinolin-2(1H)-ones through the activation of readily available transition metals, such as iron and copper. Herein, we have introduced sulfonimidoyl azide as a precursor in a Cu-catalyzed Chan-Evans-Lam reaction with aryl boronic acid for the synthesis of N-aryl sulfonimidamide. The CuCl catalyzed reactions of sulfonimidoyl azides and aryl boronic acids have been proceeded in the presence of triethylamine (Et3N) as base in methanol at room temperature under oxygen atmosphere and resulted moderate to good yields of corresponding products in very short time. The synthetic utilities of N-aryl sulfonimidamides have been established by iodination reactions and Suzuki-Miyaura coupling reactions.

Polyfunctional arenes are an important part of the chemical value chain. To improve the efficiency of their synthesis, we have investigated a multicomponent approach built upon the Catellani platform. Here, we describe a 4-component coupling of aryl iodides lacking an ortho substituent that installs 3 discrete functional groups on the arene in a single step. The process is regio- and chemoselective and uncovers remote substituent effects that have a pronounced influence over inter mediate Pd-(II) complexes. These intermediates have been a long-standing focus of the Catellani-reaction development, but persistent challenges have given rise to the well-known ortho effect. We now show that the ortho effect can be a positive element of reaction design, and that previously problematic iodides can now be used in complexity-generating transformations. In expanding the scope of the Catellani platform, we hope to provide mechanistic considerations to guide future reaction design, while also improving the environmental footprint of synthesizing polyfunctional arenes.
