Due to the continuous growth of drug resistance, finding novel fungicides with dissimilar modes of function from present fungicides for controlling downy mildew are imminent. This work is an extension of our preceding research on the original triazole sulfonamide derivatives lead compound. Triazole sulfonamide as a remarkable nitrogen-containing heterocyclic compound opposed cucumber downy mildew (CDM) develops a quite vital part in the sphere of the study of new farm chemicals. The existing report designs a certain amount of 1,2,4-triazole-1,3-disulfonamide derivatives. Hydrazones have obtained extensive attention in the field of pharmaceutical due to its unique chemical structure and remarkable activity (insecticidal, antibacterial, antifungal and herbicidal). By means of coupling numerous hydrazone with triazole sulfonyl chloride groups, 24 novel derivatives were synthesized. Spectrum analysis of LC-MS, H-1 NMR and C-13 NMR were used for characterizing these new compounds. Compared with commercial Cyazofamid using bioassays, most of these compounds displayed preferable fungicidal activities. Moreover, compounds 8q illustrated the greatest CDM resistance (EC50 = 7.776 mg/L). Field efficacy trials revealed that compound 8q fungicidal activity was higher than the purchased agrochemical Cyazofamid and Amisulbrom. Thus, the research declared that 8q displayed a great potential for the application of fungicide against CDM.

Present work demonstrates synthesis and antibacterial property of new pyrazole derivatives. A series of new Curcumin based dihydropyrazoles has been synthesized with an objective to evaluate their antibacterial property. Dihydropyrazoles analogues were synthesized using Curcumin based chalcones and differently substituted phenylhydrazine derivatives. We used the previously designed Curcumin based chalconesto react with phenylhydrazine derivatives in ethanol in a catalyst free medium to afford new dihydropyrazole derivatives. Effect of substituent on reactivity was also studied. All the synthesized pyrazole analogues were characterized using proton and carbon NMR, Mass spectroscopy and IR techniques. Effect of substituent on reactivity was explained on the basis of electronic effect generated due to groups on phenyl ring. Presence of dd (double doublet) in proton NMR spectrum of Dihydropyrazoles was also explained due to presence of optically active carbon of pyrazole ring. The synthesized library was screened for their inhibitory activity against 4 different bacterial strains 1. E. Coli (ATCC 9637), 2. Pseudomonas aeruginosa (ATCC BAA-427), 3. Staphylococcus aureus (ATCC 25923) and 4. Klebsiella pneumonia (ATCC 27736). Out of all the compounds evaluated, the compounds that exhibited IC50 value greater than 50 mu M, were considered to be inactive. We established an important SAR based on the structure dependent inhibitory potential of screened dihydropyrazoles. Two compounds 4e and 4t having nitro and benzyl substitution respectively were showing the best inhibitory potential against Gram Positive bacterial strain Staphylococcus aureus with MIC value of 1.56 mu g/ml. Compounds having Chloro and Methoxy substitution were found to be less effective against screened bacterial strains. Compounds 4a and 4b were selective towards Staphylococcus aureus species with the MIC value of 1.56 mu g/ml for each. These pyrazole analogues were not showing inhibitory potential against other screened bacterial strains.

Scaffolds containing one or more heteroatoms are ubiquitous in nature and are responsible for almost every biological processes in flora and fauna. The fact that heterocyclic cores have high propensity for biological targets, several nitrogen, oxygen, and sulfur-containing heterocyclic compounds have been reported in the last few decades. One such intriguing class is 4-thiazolidinone that exhibit diverse range of pharmacological activities. In the present study, we report synthesis, characterization, molecular docking, and anticancer evaluation of 10 new 4-thiazolidinone analogues (5-14). One of the compounds (11) was structurally characterized using single crystal X structure. Anticancer evaluation against hepatocellular carcinoma cell line (HepG2) revealed that compound (7) was the most potent (IC50=75 mu M) while other showed moderate to low activity (85-530 mu M). To underpin the druggability and possible modes of action, drug-likeness was determined and docking studies were carried out against beta-carbonic anhydrase receptor (PDB ID: 3IA). Attempts have also been made to establish a structure-activity relationship of the reported compounds.

A new group of potential pFOX inhibitors among linear methoxyphenyltriazaalkanes were designed and synthesized. Cardiotropic activity in rodent experiments was found for most of the synthesized compounds. Unprecedented triazinyl-isoxazoles were afforded via an effective cycloaddition reaction between nitrile oxides and the scarcely described 2-ethynyl-4,6-dimethoxy-1,3,5-triazine as dipolarophile. The biological evaluation of the newly synthesized compounds showed that the inhibition of human farnesyltransferase by zinc complexation could be improved with triazine-isoxazole moieties. The replacement of the isoxazole unit by a pyrrolidin-2-one was detrimental to the inhibitory activity while the pyrrolidin-2-thione derivatives conserved the biological potential. A series of tetrahydro-ss-carbolines substituted with an alkyl or acyl side chain was synthesized and screened for its antifungal activity against plant pathogenic fungi (Bipolaris oryzae, Curvularia lunata, Fusarium semitectum, and Fusarium fujikuroi). The structure activity relationship revealed that the substituent at the piperidine nitrogen plays an important role for increasing antifungal activities. In this series, 2-octyl-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole (3g) displayed potent antifungal activities with a minimum inhibitory concentration of 0.1 mu g/mL, including good inhibitory activity to the radial growth of fungus at a concentration of 100 mu g/mL compared to amphotericin B.

The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and carbonucleophiles afforded poly(arylene alkenylene)s with moderate molecular weight in good yield. The reaction involves Mizoroki-Heck coupling, olefin migration via chain walking, and addition of the carbonucleophile to the resulting pi-allylpalladium species. The polymerization with a slight excess of nucleophile with respect to diiodoarene also proceeded to give the polymer without significant decrease in molecular weight in spite of the nonstoichiometric mixture of the monomers. The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and diimide also proceeded. The base used in the reaction is critical for yield and molecular weight of the product. The reaction using NaHCO3 afforded the product with low solubility, which can be explained by the high molecular weight of the polymer and/or the strong interaction of the electron donating dimethoxyphenylene groups and electron accepting diimide groups in the polymer. In this article, we report an efficient and mild synthetic route for the construction of substituted quinazolines from functionalized 2-aminobenzophenones with various benzaldehydes usingcat. TMSOTf and hexamethyldisilazane (HMDS) under neat, metal-free and microwave irradiation conditions in which gaseous ammonia was formedin situ. This synthetic protocol provided the desired quinazolines with a broad substrate scope in good to excellent yields. Some structures were confirmed by X-ray single-crystal diffraction analysis.

A series of 2-arylidene-N-(quinolin-6-yl)hydrazine-1-carboxamides 5a-5o were synthesized and characterized. The synthesized compounds (5a-5o) were screenedin vitro against three breast cancer cell lines: SKBR3, MDA-MB-231, and MCF-7 cancer cell lines by the MTT assay. According to MTT results, compounds 5k and 5l showed better antiproliferative activities over MCF-7 cell lines with IC50 values of 8.50 and 12.51 mu M. Colony formation assay indicated 5k/5l treatment obviously inhibited the growth of MCF-7 cells and 5k/5l-induced cell cycle was arrested in the G2-M phase. Moreover, 5k/5l significantly increased the level of cleaved PARP and induced the apoptosis in MCF-7 cells. In addition, compared to Hela cells, MCF-7 cells were more sensitive to 5k/5l treatment.

Cinnamic acid and its derivatives are known for anti-tubercular activity. The present study reports the synthesis of cinnamic acid derivatives via bioisosteric replacement of terminal carboxylic acid with oxadiazole. A series of cinnamic acid derivatives (styryl oxadiazoles) were designed and synthesized in good yields by reaction of substituted cinnamic acids (2, 15a-15s) with amidoximes. The synthesized styryl oxadiazoles were evaluated in vitro for anti-tubercular activity against Mycobacterium tuberculosis (Mtb) H37Ra strain. The structure-activity relationship (SAR) study has identified several compounds with mixed anti-tubercular profiles. The compound 32 displayed potent anti-tubercular activity (IC50= 0.045 mu g/mL). Molecular docking studies on mycobacterial enoyl-ACP reductase enzyme corroborated well with the experimental findings providing a platform for structure based hit-to-lead development.
