The reaction of 2-(2,3-O-isopropylidene-5-O-trityl-beta-D-ribofuranosyl)acetonitrile (6) with isopentyl nitrite/NaH and subsequent tosylation of Na-oximino nitrile 7 gave protected 2-D-ribofuranosyl-2-(tosyloximino)acetonitriles 9alpha,beta as novel C-nucleoside precursors. Treatment of 9alpha,beta with ethyl 2-mercaptoacetate under basic conditions afforded epimeric ethyl 4-aminoisothiazole-5-carboxylate C-nucleosides 10alpha,beta. Cyclization of 10alpha,beta to the desired 3-D-ribofuranosylisothiazolo[4,5-d]pyrimidin-7(6H)-ones 14alpha,beta was accomplished by the reaction of 10alpha,beta with triethyl orthoformate and subsequent amminolysis. Deprotection of 14beta and 14alpha in 7% HCl/MeOH gave the title compound 3 and alpha-isomeric C-nucleoside 4 as monohydrochloride salt, respectively. The configuration of C-glycoside 9alpha was established by X-ray crystallography.

Enantioselective Construction of Cyclic Indolyl alpha-Amino Esters via a Friedel-Crafts Alkylation Reaction

An enantioselective Friedel-Crafts alkylation reaction of indoles with cyclic N-sulfonyl ketimino esters was developed. Under the optimized conditions using a chiral copper(II) triflate-bisoxazoline complex as the catalyst, a range of N-sulfonyl ketimino ester derivatives and indoles reacted smoothly to afford indole-containing chiral cyclic alpha-amino esters bearing tetrasubstituted alpha-stereogenic centers [3-ethoxycarbonyl-3-(3-indolyl)-2,3-dihydrobenzo[d]isothiazole 1,1-dioxides] in excellent yields and with high enantioselectivities (up to 99% ee). Pyrrole and N,N-dimethylaniline were also investigated as aromatic substrates to afford the corresponding products with good results. An asymmetric induction model was then proposed on the basis of the observed absolute configuration of the product 3-ethoxycarbonyl-3-(5-bromo-3-indolyl)-2,3-dihydrobenzo[d]isothiazole 1,1-dioxide. Synthetic transformations to convert the products into cyclic chiral N-sulfonamido alcohols and the deprotection of the sulfonamides were performed. This study provides an efficient approach to chiral alpha-tetrasubstituted indolic alpha-amino acids as potential building blocks for peptides and biologically active molecules.

Differences in kinetics of structurally related competitive GABA(A) receptor antagonists

Previously, 4-alkyl and 4-aryl substituted analogues of the low-efficacy partial GABA(A) receptor agonist 5-(4-piperidyl)-3-isothiazole (4-PIOL) have been identified as competitive GABAA receptor antagonists. These structurally related competitive antagonists show marked differences in their kinetic properties. The kinetics of 20 4-alkyl and 4-aryl substituted analogues of 4-PIOL, two 4-arylalkyl substituted 3-isothiazolol analogues and the classical GABAA receptor antagonist SR95531 was studied in cultured cerebral cortical neurons using whole-cell patch-clamp techniques. The kinetics of the antagonists was studied indirectly by measuring the changes in the response of the full GABAA receptor agonist isoguvacine (IGU) induced by concurrent application of an antagonist. When added, the majority of the antagonists did not affect the rate of deactivation of the IGU-induced responses. When removed, however, the majority of the antagonists slowed the reactivation phase of IGU implying that the dissociation of the antagonist from the GABAA receptor is the rate-limiting step. Surprisingly, the functional off-rates of the antagonists seemed to correlate better with the lipophilicity of the compounds than with the affinity and potency. This suggests that the dissociation of the tested antagonists from the GABAA receptor is restricted by lipophilic interactions, perhaps with the aromatic amino acids surrounding the GABA binding site.
