Members of two novel families of spiro(oxazolidinediones) have been prepared. Spiro[1H-isoindole-1,5′-oxazolidine]-2′, 3(2H),4′-triones (4) were synthesized from the corresponding 1-hydroxy-1-carboxyethylisoindol-3-one (7) or (10) or their primary amide derivatives (14). In turn 7 and 10 were prepared from 2-bromobenzoic acid amides (6) and (9). or from N-alkylated isoquinolin-1, 3, 4(2H)-triones (12). Spiro[1, 2-benzisothiazole-3(2H), 5′-oxazolidine]-2′, 4′-dione 1, 1-dioxides (5) were constructed from the appropriate 2,3-dihydro-3-hydroxy-1,2-benzisothiazole-3-carboxamide 1,1-dioxide (17). The immediate precursors to 14, ethyl esters (16) and (19), were prepared from benzenesulfonamide (15), or from 2-bromobenzenesulfonamide (18).

Isothiazolinones are used in cosmetic and as chemical additives for occupational and industrial usage due to their bacteriostatic and fungiostatic activity. Despite their effectiveness as biocides, isothiazolinones are strong sensitizers, producing skin irritations and allergies and may pose ecotoxicological hazards. Therefore, their use is restricted by EU legislation. Considering the relevance and importance of isothiazolinone biocides, the present review describes the state-of-the-art knowledge regarding their synthesis, antibacterial components, toxicity (including structure-activity-toxicity relationships) outlines, and (photo)chemical stability. Due to the increasing prevalence and impact of isothiazolinones in consumer’s health, analytical methods for the identification and determination of this type of biocides were also discussed.

Isothiazole dioxides have been shown to inhibit Trypanosoma brucei protein. farnesyltransferase (PFTase) in isolated enzyme, but elicited only a minor effect on mammalian PFTase. In the present study we have evaluated the effect of 3-diethylamino-4-(4-methoxyphenyl)-isothiazole 1,1-dioxides with different substituents at C5, on rat PFTase and protein geranylgeranyltransferase-I (PGGTase-1) with the final aims to improve the potency against mammalian PFTase and to identify new compounds with antiproliferative properties. For these purposes, in vitro and cell culture models have been utilized. The results showed that isothiazole dioxides with C4-C5 double bond and sulfaryl substituted at the C5 position but none of the dihydro-derivatives, were able to inhibit in vitro PFTase in a concentration dependent manner (IC50 ranging from 8.56 to 1015 mu M). Among those, compound 6n (C5; methyl-S) displayed 500-fold higher inhibitory potency on PFTase than PGGTase-1. Compound 6n was shown to affect rat smooth muscle cell (SMC) proliferation at concentrations similar (IC50 = 61.4 mu M) to those required to inhibit [H-3]-farnesol incorporation into cellular proteins (-44.1% at 100 mu M). Finally, compound 6n interferes with rat SMC proliferation by blocking the progression of G0/G1 phase without inducing apoptosis, as assessed by [H-3]-thymidine incorporation assay and flow cytometry analysis. Taken together, we described a new PFTase inhibitor containing the isothiazole dioxide moiety that affects mammalian protein farnesylation and SMC proliferation by inhibiting G0/ G1 phase of the cell cycle. Rational design of diketopyrrolopyrrole-based multifunctional materials for organic light-emitting diodes and organic solar cells

A series of donor-acceptor diketopyrrolopyrrole derivatives has been investigated to reveal their optical, electronic, and charge transport properties for applications in organic light-emitting diodes (OLEDs) and organic solar cells (OSCs). The calculated results show that their optical, electronic, and charge transport properties are affected by the different end groups. The introduction of the 2,5-diphenylthiophene, benzo[c]isothiazole, benzo[c]thiophene, benzo[c]thiophene, 2,4-dihydrobenzo[d]thiazole, and thieno[3,4-b][1,4] dioxine end groups can broaden absorption spectrum and improve charge transport property of the designed compounds. Our results suggest that the designed compounds can serve as donor materials for OSCs and luminescent materials for OLEDs. In addition, the mobility of the designed compounds is also investigated. They are expected to be promising candidates for hole and electron transport materials. On the basis of investigated results, we proposed a rational way for the design of multifunctional materials for OLED and OSC applications.
