A novel palladium(II)/copper(II)-catalyzed sulfidation of the C-H bond in electron-rich arenes and in pentafluorobenzene with disulfides was developed. This catalytic system can be used to efficiently produce various types of either unsymmetrical aryl sulfides or alkyl aryl sulfides. The present protocol could also be applied to the direct preparation of unsymmetrical aryl selenides via C-H selenation.

Two series of polymethoxychalcones and polymethoxyflavones, including the natural products 2-hydroxy-3,4,5,4,6-pentamethoxychalcone (8c), 5,6,7,8,3,4,5-heptamethoxyflavone (6), 5,7,3,4,5-pentamethoxyflavone (9c), 3-hydroxy-5,6,7,8,3,4,5-heptamethoxyflavone (7), and 3-hydroxy-5,7,3,4,5-pentamethoxyflavone (10), were synthesized. The antiproliferative activity in vitro was evaluated against a panel of three human cancer cell lines (HeLa, HCC1954, and SK-OV-3) by the cell counting kit-8 (CCK-8) assay. The results showed that most of the synthetic compounds exhibited moderate to potent antiproliferative activities. Some compounds displayed equal or higher potential than the positive control drug cisplatin. In particular, compounds 4c, 4e, 8a, and 9a possess IC50 values equal to or below 10 M and are worthy of further investigation.

A photocatalyst-free and mild visible light photochemical procedure for C(sp(3))-H arylation of amides is described. The reaction proceeds via an electron donor-acceptor (EDA) complex between an electron-rich arene substrate and electron-poor persulfate oxidant. C(sp(3))-H arylation of the amide occurs selectively with the most electron-rich arene of the substrate. Mechanistic studies corroborate the reaction taking place in a solvent cage holding components in a close proximity.

We developed a mild aromatic halogenation reaction using a combination of N-halosuccinimide and PhSSiMe3 or PhSSPh. Less reactive aromatic compounds, such as methyl 4-methoxybenzoate, were brominated with PhSSiMe3 or PhSSPh and N-bromosuccinimide in high yields. No reaction was observed in the absence of PhSSiMe3 or PhSSPh. This method is also applicable to chlorination reactions using N-chlorosuccinimide and PhSSPh.

Electron-rich arenes are oxidatively photochlorinated in the presence of catalytic amounts of bromide ions, visible light, and 4CzIPN as organic photoredox catalyst. The substrates are brominated in situ in a first photoredox-catalyzed oxidation step, followed by a photocatalyzed ipso-chlorination, yielding the target compounds in high ortho/para regioselectivity. Highly selective direct monofluorination of indoles and arenes was developed through an approach that allows site-specific solubility of substrate and fluorine source in the micelle. This approach was highly selective for a broad range of substrates with excellent functional group tolerance. Differences in binding constant and solubility of indoles and arenes in the micelle allowed the fine-tuning of selectivity. Control experiments suggested a radical pathway and provided insight into the role of micelles of the environmentally benign amphiphile PS-750-M. Dynamic light scattering experiments strongly indicated the site-specific solubility of the substrate and fluorine source. The methodology was successfully adapted to gram scale, and the E-factor established from a recycle study indicated that the process is environmentally responsible and sustainable.

Purpose: Recombinant human growth hormone (rhGH) has been used to improve growth in children with Noonan syndrome (NS). This study aimed to investigate the efficacy of rhGH therapy in Korean children with NS. Methods: Seventeen prepubertal children (10 boys, 7 girls) with NS who received rhGH therapy for at least 3 years between 2008 and 2017 were included. To compare the response, age- and sex-matched children with GH deficiency (GHD; n=31) were included. Height and growth velocity before and during treatment were analyzed. Results: The mean age of NS patients was 6.34 +/- 2.32 years. After treatment, the height standard deviation score (SDS) increased from -2.93 +/- 0.81 to -1.51 +/- 1.00 in patients with NS and from -2.45 +/- 0.42 to -1.09 +/- 0.47 in patients with GHD. There were no significant differences in growth velocity or change in height SDS between patients with NS and GHD. Growth velocity in the first year of treatment was higher in patients with PTPN11 mutations than those without PTPN11 mutations, but the change in height SDS was not significantly different between those 2 groups. Conclusion: rhGH therapy can increase linear growth in prepubertal children with NS. The growth response between patients with NS and patients with GHD was not significantly different. Furthermore, we observed that lower doses of growth hormone have a similar effect on height compared to previous studies in patients with NS. Our study indicates that rhGH treatment is useful for growth promotion.
