This process provides a direct approach to alpha-carbonyloxy esters with the achievement of formation of three C-O bonds. The protocol is featured with readily available starting materials and broad substrate scope. Control reactions and isotope-labeling reactions were conducted to elucidate a plausible mechanism.

This report describes the formation of value-added acyl fluorides by means of palladium-catalyzed acyl-exchange reactions between acyl fluorides and acid anhydrides. This method allows using a simple and commercially available acyl fluoride, benzoyl fluoride, as the fluoride source for the easy and efficient preparation of a variety of more complex acyl fluorides. The results of this study suggest that this reaction proceeds via a reversible acyl C-F bond cleavage/formation at the palladium center.

The results of a comparative study of phosphine-containing and phosphine-free catalytic systems of the Heck reaction using aromatic carboxylic anhydrides as arylating agents are presented. It was demonstrated that the patterns of diff erential selectivity of the reaction under competition of two aromatic anhydrides or two alkenes are independent of the presence of a tertiary phosphine additive in the system. It was established that palladium complexes with no phosphine ligands in their coordination sphere are catalytically active at the step of activation of aromatic carboxylic anhydride and alkene. The patterns of diff erential selectivity for regioisomers of arylated products provide the evidence of the participation of phosphine-containing anionic palladium complexes in the regioselectivity-determining step of the catalytic cycle. The data obtained are in agreement with the phosphine involvement in the catalyst transformations proceeding outside the main catalytic cycle.

Redox modulators have been developed as an attractive approach to treat cancer. Herein, we report the synthesis, identification, and biological evaluation of a quinazolinedione reactive oxygen species (ROS) inducer, QD394, with significant cytotoxicity in pancreatic cancer cells. QD394 shows a transcriptomic profile remarkably similar to napabucasin, a cancer stemness inhibitor. Both small molecules inhibit STAT3 phosphorylation, increase cellular ROS, and decrease the GSH/GSSG ratio. Moreover, QD394 causes an iron- and ROS-dependent, GPX4 mediated cell death, suggesting ferroptosis as a major mechanism. Importantly, QD394 decreases the expression of LRPPRC and PNPT1, two proteins involved in mitochondrial RNA catabolic processes and both negatively correlated with the overall survival of pancreatic cancer patients. Pharmacokinetics-guided lead optimization resulted in the derivative QD394-Me, which showed improved plasma stability and reduced toxicity in mice compared to QD394. Overall, QD394 and QD394-Me represent novel ROS-inducing drug-like compounds warranting further development for the treatment of pancreatic cancer.

Herein, we have introduced sulfonimidoyl azide as a precursor in a Cu-catalyzed Chan-Evans-Lam reaction with aryl boronic acid for the synthesis of N-aryl sulfonimidamide. The CuCl catalyzed reactions of sulfonimidoyl azides and aryl boronic acids have been proceeded in the presence of triethylamine (Et3N) as base in methanol at room temperature under oxygen atmosphere and resulted moderate to good yields of corresponding products in very short time. The synthetic utilities of N-aryl sulfonimidamides have been established by iodination reactions and Suzuki-Miyaura coupling reactions.
