Hydroxynitrile lyase (HNL) catalyzed enantioselective C-C bond formation is an efficient approach to synthesize chiral cyanohydrins which are important building blocks in the synthesis of a number of fine chemicals, agrochemicals and pharmaceuticals. Immobilization of HNL is known to provide robustness, reusability and in some cases also enhances activity and selectivity. We optimized the preparation of immobilization of Baliospermium montanum HNL (BmHNL) by cross linking enzyme aggregate (CLEA) method and characterized it by SEM. Optimization of biocatalytic parameters was performed to obtain highest % conversion and ee of (S)-mandelonitrile from benzaldehyde using CLEA-BmHNL. The optimized reaction parameters were: 20 min of reaction time, 7 U of CLEA-BmHNL, 1.2 mM substrate, and 300 mM citrate buffer pH 4.2, that synthesized (S)-mandelonitrile in 99% ee and 60% conversion. Addition of organic solvent in CLEA-BmHNL biocatalysis did not improve in % ee or conversion of product unlike other CLEA-HNLs. CLEA-BmHNL could be successfully reused for eight consecutive cycles without loss of conversion or product formation and five cycles with a little loss in enantioselectivity. Eleven different chiral cyanohydrins were synthesized under optimal biocatalytic conditions in up to 99% ee and 59% conversion, however the % conversion and ee varied for different products. CLEA-BmHNL has improved the enantioselectivity of (S)mandelonitrile synthesis compared to the use of purified BmHNL. Nine aldehydes not tested earlier with BmHNL were converted into their corresponding (S)-cyanohydrins for the first time using CLEA-BmHNL. Among the eleven (S)-cyanohydrins syntheses reported here, eight of them have not been synthesized by any CLEA-HNL. Overall, this study showed preparation, characterization of a stable, robust and recyclable biocatalyst i.e. CLEA-BmHNL and its biocatalytic application in the synthesis of different (S)-aromatic cyanohydrins.

In this study, two new copper (II) carboxylate complexes, [Cu(4-mba)(2) (tmeda)](1) and [Cu(3mba)(2)(H2O)(2)(tmeda)](2) (4-mbaH = 4-methylbenzoic acid, 3-mbaH = 3-methylbenzoic acid, tmeda = N,N,N’,N’-tetramethylethyleneamine) were synthesized. The structures and properties of the complexes were determined by X-ray single crystal diffraction method and FT-IR spectroscopy. While the crystal packing of the complex 1 is stabilized by C-H center dot center dot center dot O hydrogen bonds, in the complex 2, the molecules are connected to each other by C-H center dot center dot center dot O and O-H center dot center dot center dot O hydrogen bonds. The optimized structures were formed using density functional theory (DFT) at the B3LYP level within the 6-311G(d,p) and LAN2DZ basis sets. Their Hirshfeld surface analyses were performed for the detailed investigation of all the contacts involved in the crystal packing. 2D fingerprint plots and Hirshfeld surface analyses signify important interactions in the crystal packing [H center dot center dot center dot H (64.2%), C center dot center dot center dot H/H center dot center dot center dot C (21.1%) and O center dot center dot center dot H/H center dot center dot center dot O (14.7%) contacts for [Cu(4-mba) 2(tmeda)], while [Cu(3-mba)(2)(H2O)(2)(tmeda)] showed H center dot center dot center dot H (67.3%), C center dot center dot center dot H/H center dot center dot center dot C (21.8%) and O center dot center dot center dot H/H center dot center dot center dot O (10.9%) contacts]. This operationally simple alkylation proceeds under ambient conditions with high efficiency, regioselectivity, and an exceptionally broad scope of arene nucleophiles. A key feature of this alkylation is the role associated with the silver(I) salt counteranions liberated during the reaction. Taking advantage of a phase-transfer counteranion/Bronsted acid pair mechanism, we also report a catalytic enantioselective example of the reaction.

A one-pot, multicomponent-type, solvent-free Pd(II)-catalyzed direct beta-C-H activation/arylation of carboxamides involving anhydrides as substrates via in situ installation of directing group (DG) is reported. Typically, the DG-assisted beta-C-H activation/arylation of carboxamides is a two-step process comprising the installation of DG and Pd(II)-catalyzed C-H arylation. We attempted a multicomponent-type reaction comprising an anhydride, a DG (e.g. 8-aminoquinoline), an aryl iodide in the presence of the Pd(II) catalyst and an appropriate additive. Different anhydrides, DGs, aryl iodides, catalysts and additives were screened to reveal the scope of this multicomponent-type C-H arylation reaction process and various beta-C-H arylated carboxamides were obtained in satisfactory to good yields. The Rh-catalyzed C-H alkylation of benzylamine derivatives with unactivated 1-alkenes that proceeds via a picolinamide directing group is reported. The crucial role of an acid additive in this transformation is confirmed. Aromatic acids showed high linear selectivity, and aliphatic acids provided branched alkylation products as the major product. The reaction has a broad scope for benzylamines and alkenes. Deuterium labeling experiments suggest that a Rh-carbene intermediate is involved in the case of linear product formation. A different reaction pathway, however, appears to be involved in the case of branched alkylation products, and this pathway also appeared to be a minor pathway in linear-selective reactions.

We developed a mild aromatic halogenation reaction using a combination of N-halosuccinimide and PhSSiMe3 or PhSSPh. Less reactive aromatic compounds, such as methyl 4-methoxybenzoate, were brominated with PhSSiMe3 or PhSSPh and N-bromosuccinimide in high yields. Herein, we describe two novel strategies for the synthesis of esters, as achieved under high-speed ball-milling (HSBM) conditions at room temperature. In the presence of I-2 and KH2PO2, the reactions afford the desired esterification derivatives in 45% to 91% yields within 20 min of grinding. Meanwhile, using KI and P(OEt)(3), esterification products can be obtained in 24% to 85% yields after 60 min of grinding. In addition, the I-2/KH2PO2 protocol was successfully extended to the late-stage diversification of natural products showing the robustness of this useful approach. Further application of this method in the synthesis of inositol nicotinate was also discussed.

We report the preparation of lithium-salt-free KDA (potassium diisopropylamide; 0.6 m in hexane) complexed with TMEDA (N,N,N ‘,N ‘-tetramethylethylenediamine) and its use for the flow-metalation of (hetero)arenes between -78 degrees C and 25 degrees C with reaction times between 0.2 s and 24 s and a combined flow rate of 10 mL min(-1) using a commercial flow setup. The resulting potassium organometallics react instantaneously with various electrophiles, such as ketones, aldehydes, alkyl and allylic halides, disulfides, Weinreb amides, and Me3SiCl, affording functionalized (hetero)arenes in high yields. This flow procedure is successfully extended to the lateral metalation of methyl-substituted arenes and heteroaromatics, resulting in the formation of various benzylic potassium organometallics. A metalation scale-up was possible without further optimization.

Background Isophthalic acid (IPA) is commercially produced using the Co-Mn-Br catalyst system, which suffers from the shortcomings of the discharge of CH3Br and corrosion of equipment. It is necessary to develop a technology to avoid the pollution caused by bromide. The production of IPA from the oxidation ofm-xylene (MX) by air is realized under catalysis with H3PW12O40(HPW) loaded on carbon and cobalt. Results Tartaric acid has been used to improve the catalytic activity of the HPW@C catalyst. Experiments indicate that the best modification condition is calcining the carbon at 450 degrees C for 4 h after being soaked in a 2.0 mol L(-1)tartaric acid solution for 10 h. Surface characterization reveals that the tartaric acid modification leads to an increase in the acid groups and a reduction in the basic groups on the carbon surface. Conclusions The MX conversion obtained using the HPW@C catalyst prepared from modified carbon is 6.77% over that obtained using the HPW@C catalyst prepared from the original carbon. The IPA generated using the former is 71.1% over that generated using the latter. The catalytic activity of the HPW@C catalyst relies on its surface chemical characteristics and physical properties. A novel silica-based organic-inorganic hybrid material namely N,N,N ‘,N ‘-tetramethyl-N-(silica-n-propyl)-N ‘-sulfonic acid-ethylenediaminium chloride/mesylate ([TSSECM]) was prepared, and characterized by EDS (energy-dispersive X-ray spectroscopy), FE-SEM (field emission scanning electron microscopy), FT-IR, XRD (X-ray diffraction), TGA (thermal gravimetric analysis) and adsorption/desorption porosimetry analyses. The sizes of most particles of [TSSECM] were less than 100 nm (i.e. they were nanomaterials), and some particles had sizes more than 100 nm (up to 185 nm). The hybrid material was applied as a highly efficacious, recyclable and dual-functional catalyst for the one-pot multi-component reaction of arylaldehydes, dimedone and 6-amino-1,3-dimethyluracil under solvent-free conditions to give pyrimido[4,5-b]quinolines.
