Copper (II) immobilized on L-arginine-beta-cyclodextrin-functionalized magnetite nanoparticles (nano-Fe3O4@L-arginine-CD-Cu(II)) were successfully synthesized and fully characterized using FT-IR, XRD, SEM, EDX, ICP, TGA and VSM techniques. The catalytic activity of these magnetically retrievable nanoparticles was evaluated in the direct oxidation of primary alcohols and benzyl halides to acids in neat conditions that was observed to proceed well and products were obtained in good yields. In addition to showing good catalytic activity, the magnetic catalyst is easy to synthesize and can be recycled at least five times with little loss in activity. (C) 2020 Elsevier B.V. Chalcone based structures have many interesting biological properties like anti-malarial, anticancer, antiviral, antibacterial, antifungal, anti-hyperglycaemic and photo-cytotoxicity activities. In this study, the novel chalcone derivatives (1a-11) have been synthesized and investigated for antimicrobial activities. The MIC results of antimicrobial activities reveal that one of the chalcone derivative (2-Br- and 4OCH(3)- group) against specific bacteria are quite significant. 3D QSAR, three dimensional quantitative structure activity is one of the most powerful technique which involves the quantitative relationship between the biological activity of a set of compounds and their three dimensional structural properties, using statistical correlation methods. Pharmacophore modelling and molecular docking play an important role in drug design. To elucidate the relationship between structures and its activity, field-based 3D QSAR analysis has been carried out of novel chalcone derivatives. Structural properties such as electrostatic, hydrophobic, aromatic, and hydrogen-bond donor and acceptor have been worked out for novel chalcone derivatives. To investigate the influence of drugstore, chalcone derivatives (1a-11) have docked with receptor 210K and results displayed the highest Gscore of ligand If (3-OCH2C6H5-4-OCH3- group).

Cleavage of C-O bonds in lignin can afford the renewable aryl sources for fine chemicals. However, the high bond energies of these C-O bonds, especially the 4-O-5-type diaryl ether C-O bonds (similar to 314 kJ/mol) make the cleavage very challenging. Here, we report visible-light photoredox-catalyzed C-O bond cleavage of diaryl ethers by an acidolysis with an aryl carboxylic acid and a following one-pot hydrolysis. Two molecules of phenols are obtained from one molecule of diaryl ether at room temperature. The aryl carboxylic acid used for the acidolysis can be recovered. The key to success of the acidolysis is merging visible-light photoredox catalysis using an acridinium photocatalyst and Lewis acid catalysis using Cu (TMHD)(2). Preliminary mechanistic studies indicate that the catalytic cycle occurs via a rare selective electrophilic attack of the generated aryl carboxylic radical on the electron-rich aryl ring of the diphenyl ether. This transformation is applied to a gram-scale reaction and the model of 4-O-5 lignin linkages.

We have synthesized 50 benzimidazole (BMZ) derivatives with 1,2-phenylenediamines and aromatic aldehydes under mild oxidation conditions by using inexpensive, nontoxic inorganic salt sodium metabisulfite in a one-pot condensation reaction and screened their ability to interfere with Zika virus (ZIKV) infection utilizing a cell-based phenotypic assay. Seven BMZs inhibited an African ZIKV strain with a selectivity index (SI=CC50/EC50) of 9-37. Structure-activity relationship analysis demonstrated that substitution at the C-2, N-1, and C-5 positions of the BMZ ring were important for anti-ZIKV activity. The hybrid structure of BMZ and naphthalene rings was a structural feature responsible for the high anti-ZIKV activity. Importantly, BMZs inhibited ZIKV in human neural stem cells, a physiologically relevant system considering the severe congenital anomalies, like microcephaly, caused by ZIKV infection. Compound 39 displayed the highest antiviral efficacy against the African ZIKV strain in Huh-7 (SI>37) and neural stem cells (SI=12). Compound 35 possessed the highest activity in Vero cells (SI=115). Together, our data indicate that BMZs derivatives have to be considered for the development of ZIKV therapeutic interventions.

A novel series of zileuton-hydroxycinnamic acid hybrids were synthesized and screened as 5-lipoxygenase (5-LO) inhibitors in stimulated HEK293 cells and polymorphonuclear leukocytes (PMNL). Zileuton’s (1) benzo[b]thiophene and hydroxyurea subunits combined with hydroxycinnamic acid esters’ ester linkage and phenolic acid moieties were investigated. Compound 28, bearing zileuton’s (1) benzo[b]thiophene and sinapic acid phenethyl ester’s (2) alpha,beta-unsaturated phenolic acid moiety 28, was shown to be equipotent to zileuton (1), the only clinically approved 5-LO inhibitor, in stimulated HEK293 cells. Compound 28 was three times as active as zileuton (1) for the inhibition of 5-LO in PMNL. Compound 37, bearing the same sinapic acid (3,5-dimethoxy-4-hydroxy substitution) moiety as 28, combined with zileuton’s (1) hydroxyurea subunit was inactive. This result shows that the zileuton’s (1) benzo[b]thiophene moiety is essential for the inhibition of 5-LO product biosynthesis with our hydrids. Unlike zileuton (1), Compound 28 formed two pi-pi interactions with Phe177 and Phe421 as predicted when docked into 5-LO. Compound 28 was the only docked ligand that showed a pi-pi interaction with Phe177 which may play a part in product specificity as reported.
