An operational approach for synthesis of eleven substituted 4H-chromene derivatives by one-pot, three-component reaction of aromatic aldehyde, methyl cyanoacetate and cyclic diketone through ultrasound irradiation, in the presence of aqueous ethanol under catalyst-free condition is reported. Different spectral methods, including H-1, C-13 and N-15 NMR and HRMS were utilized to characterize the new molecules. Benefits of the eco-friendly method are fast reaction (10 min), excellent yields (92-98%), no column chromatography and no byproducts. Reaction offers 94% atom economy and 100% carbon capture. Highly selective direct monofluorination of indoles and arenes was developed through an approach that allows site-specific solubility of substrate and fluorine source in the micelle. This approach was highly selective for a broad range of substrates with excellent functional group tolerance. Differences in binding constant and solubility of indoles and arenes in the micelle allowed the fine-tuning of selectivity. Control experiments suggested a radical pathway and provided insight into the role of micelles of the environmentally benign amphiphile PS-750-M. Dynamic light scattering experiments strongly indicated the site-specific solubility of the substrate and fluorine source. The methodology was successfully adapted to gram scale, and the E-factor established from a recycle study indicated that the process is environmentally responsible and sustainable.

This contribution describes sulfoximine (SFI) derivatives as a novel type of acceptor for efficient thermally activated delayed fluorescence (TADF) emitters. SFI presents a unique advantage by facile introduction of versatile functionalities on its secondary amine, which allows delicate manipulation of TADF properties. In a proof of principle study, three green/yellowish green TADF emitters, namely DMAC-SFIP, DMAC-SFIC and DMAC-SFIS were developed with different electron-withdrawing N-substituents based on simple donor-acceptor (D-A) typed structure. A look into the molecular conformations indicates the existence of different types of weak intramolecular interactions to suppress nonradiative decay. More importantly, effective mixing between charge transfer (CT) and locally excited (LE) triplet states is observed for DMAC-SFIP, DMAC-SFIC due to suitable electron-deficiency in the SFI segment, resulting in acceleration of the reverse intersystem crossing (RISC) rate. Consequently, these new TADF emitters render excellent organic light-emitting diode (OLED) performances with external quantum efficiencies (EQEs) up to 20.4% and small efficiency roll-off.

The palladium-catalyzed selective activation of C-H bonds is of great importance for the construction of diverse bioactive molecules. Herein, monodisperse CuPd alloy nanoparticles supported on reduced graphene oxide have been used as high efficient and recyclable catalyst for C(sp(3))-H acyloxylation of 8-methylquinolines in good yields under mild conditions. The developed catalytic system proceeds with low palladium loading, good catalyst recyclability, high step and atom efficiency, excellent chemo and regioselectivity, good substrate and functional group compatibility, and applicable for large scale up synthesis. Preliminary studies were carried out to gain insights into the heterogeneous catalysis mechanism.

The chlorination of a bioactive compound can change its physiological properties and improve its pharmacokinetic and pharmacological profiles. It therefore has been an important strategy for drug discovery and development. However, the direct aromatic chlorination of complex bioactive molecules is too difficult to be practical. In fact, many functional groups such as hydroxyls, amines, amides or carboxylic acids may strongly restrain the reactivity of Cl+ by forming a halogen bond. Here we report a highly efficient aromatic chlorination of arenes that is catalysed by dimethyl sulfoxide with N-chlorosuccinimide as the chloro source. The mild conditions, easy-availability and stability of the catalyst and reagents, as well as good functional-group tolerance, showed the approach to be a versatile protocol for the late-stage aromatic chlorination of complex natural products, drugs and peptides. The multi-gram experiment and low-cost of N-chlorosuccinimide and dimethyl sulfoxide shows great potential for drug discovery and development in industrial applications. Late-stage aromatic chlorination of active pharmaceutical ingredients has enormous potential in drug discovery yet still features limited applicability due to issues of functional-group tolerance. Now, dimethyl sulfoxide is reported as catalyst for the chlorination of a diverse family of bioactive molecules in combination with N-chlorosuccinimide.

Aurora and polo-like kinases control the G2/M phase in cell mitosis, which are both considered as crucial targets for cancer cell proliferations. Here, naphthalene-based Aurora/PLK coinhibitors as leading compounds were designed through in silico approach, and a total of 36 derivatives were synthesized. One candidate (AAPK-25) was selected under in vitro cell based high throughput screening with an IC50 value = 0.4 mu M to human colon cancer cell HCT-116. A kinome scan assay showed that AAPK-25 was remarkably selective to both Aurora and PLK families. The relevant genome pathways were also depicted by microarray based gene expression analysis. Furthermore, validated from a set of in vitro and in vivo studies, AAPK-25 significantly inhibited the development of the colon cancer growth and prolonged the median survival time at the end of the administration (p < 0.05).
