Herein, we report the synthesis of the Man(9)epitope with both alpha and beta configurations at the reducing end, and subsequent evaluation of the impact of this configuration on binding to natural receptor of high-mannose, DC-SIGN. Using fluorescence polarization assays, we demonstrate that both anomers bind to DC-SIGN with comparable affinity. These relevant results therefore indicate that the more synthetically-accesible Man(9)alpha epitope may be deployed as ligand for DC-SIGN in bothin vitroandin vivobiological assays.

The reaction of zirconium salts with meso-tetra(4-carboxyphenyl)porphyrin (TCPP) in the presence of different modulators results in the formation of a diverse set of metal-organic frameworks (MOFs), each displaying distinct crystalline topologies. However, the synthesis of phase-pure crystalline frameworks remains challenging due to the concurrent formation of different polymorphs. The acidity and concentration of the modulator greatly influence the outcome of the MOF synthesis. By systematically varying these two parameters, selective framework formation can be achieved. In the present study, we aimed to elucidate the effect of modulator on the synthesis of zirconium-based TCPP MOFs. With the help of powder X-ray diffraction and scanning electron microscopy, modulator candidates and the optimal synthetic conditions yielding phase-pure PCN-222, PCN-223, and MOF-525 were identified. H-1 nuclear magnetic resonance analysis, thermogravimetric analysis, and N-2 gas sorption measurements were performed on select MOFs to gain insight into the relationship between their defectivity and modulator properties.

Metal single-atom catalysts (SACs) promise great rewards in terms of metal atom efficiency. However, the requirement of particular conditions and supports for their synthesis, together with the need of solvents and additives for catalytic implementation, often precludes their use under industrially viable conditions. Here, we show that palladium single atoms are spontaneously formed after dissolving tiny amounts of palladium salts in neat benzyl alcohols, to catalyze their direct aerobic oxidation to benzoic acids without ligands, additives, or solvents. With this result in hand, the gram-scale preparation and stabilization of Pd SACs within the functional channels of a novel methyl-cysteine-based metal-organic framework (MOF) was accomplished, to give a robust and crystalline solid catalyst fully characterized with the help of single-crystal X-ray diffraction (SCXRD). These results illustrate the advantages of metal speciation in ligand-free homogeneous organic reactions and the translation into solid catalysts for potential industrial implementation.

In this work, Cyrene (TM), a bio-based green solvent, has been transformed into its methacrylic derivative (m-Cyrene) for the first time. Polymerization of the newly-synthesized monomer was studied using different polymerization techniques such as bulk, solution and emulsion polymerization. Compared to solution polymerization, emulsion polymerization was found to produce higher yields and molecular weights. Cyrene (TM), along with other green solvents, was used as a polymerization solvent and was found to be the most effective (as a solvent) with the highest yield and molecular weight. The new Cyrene (TM) monomer was found to undergo rapid polymerization compared to isobornyl methacrylate (IBMA), another bio-derived monomer with a close structural similarity, and the highest polymerization rate was obtained in Cyrene (TM) as the polymerization solvent media. A reactivity ratio calculation also showed m-Cyrene to be more reactive than IBMA. The homopolymer derived from emulsion polymerization showed high glass transition temperature (192 degrees C) and thermal stability (up to ca. 266 degrees C). Cytotoxicity testing confirmed the non-toxic nature of the bio-derived monomer, making it a green bio-derived methacrylic monomer for synthesizing polymers, where high thermal stability and mechanical properties are required.

The results of a comparative study of phosphine-containing and phosphine-free catalytic systems of the Heck reaction using aromatic carboxylic anhydrides as arylating agents are presented. It was demonstrated that the patterns of diff erential selectivity of the reaction under competition of two aromatic anhydrides or two alkenes are independent of the presence of a tertiary phosphine additive in the system. It was established that palladium complexes with no phosphine ligands in their coordination sphere are catalytically active at the step of activation of aromatic carboxylic anhydride and alkene. The patterns of diff erential selectivity for regioisomers of arylated products provide the evidence of the participation of phosphine-containing anionic palladium complexes in the regioselectivity-determining step of the catalytic cycle. The data obtained are in agreement with the phosphine involvement in the catalyst transformations proceeding outside the main catalytic cycle.

Imidazole-based epidermal growth factor receptor (EGFR) inhibitors were computationally designed and synthesized.
