We successfully demonstrated that Friedel-Crafts-type allylation of phenol derivatives and allylic phosphates is catalyzed by the AgTFA/trimethylsilyl cyanide (Me3SiCN) and Pd(OAc)(2)/Me3SiCN combined systems to afford the C-allylated product in a highly regioselective manner. The corresponding silyl cyanometallates generated in situ are proposed to be the active catalytic species. Lewis acidity of the reversibly formed ion pairs is appropriately regulated for this reaction. The para-allylated anisole and phenol derivatives are selectively obtained. The para-substituted ones are converted to the ortho-allylated products. The reactivity of the catalytic systems is strongly dependent on the electronic nature of both electrophile and nucleophile. Substitution of an aromatic ring on the allylic phosphate is essential for the reaction. Thus, the competitive reaction of a 1 : 1 mixture of cinnamyl and simple allyl phosphates affords only the cinnamyl-substituted product.

It remains challenging to achieve the selective cleavage of C-C bonds in lignin or lignin model compounds to produce aromatic products in high yield and selectivity. We have developed a redox-neutral photocatalytic strategy to accomplish this goal in both beta-O-4 and beta-1 lignin models at room temperature (RT) via proton-coupled electron transfer (PCET) process without any pretreatments of substrate, by adjusting the alkalinity of base to obtain a lignin models/base PCET pair with a bond dissociation free energy close to 102 kcal/mol. Without breaking down C-beta-C-gamma bond and any C-O bonds, this PCET method is 100% atom economy and produces exclusive C-alpha-C-beta bond cleavage products, such as benzaldehydes (up to 97%) and phenyl ethers (up to 96%), in high to excellent yields and selectivities. Preliminary studies indicated that the PCET strategy is also effective for the depolymerization of native lignin at RT, thus providing significantly important foundation to the depolymerization of lignin. The metal-free activation of C(sp(3))-H bonds to value-added products is of paramount importance in organic synthesis. We report the use of the commercially available organic dye 2,4,6-triphenylpyrylium tetrafluoroborate (TPP) for the conversion of methylarenes to the corresponding aryl nitriles via a photocatalytic process. Applying this methodology, a variety of cyanobenzenes have been synthesized in good to excellent yield under metal- and cyanide-free conditions. We demonstrate the scope of the method with over 50 examples including late-stage functionalization of drug molecules (celecoxib) and complex structures such as l-menthol, amino acids, and cholesterol derivatives. Furthermore, the presented synthetic protocol is applicable for gram-scale reactions. In addition to methylarenes, selected examples for the cyanation of aldehydes, alcohols and oximes are demonstrated as well. Detailed mechanistic investigations have been carried out using time-resolved luminescence quenching studies, control experiments, and NMR spectroscopy as well as kinetic studies, all supporting the proposed catalytic cycle.

A novel series of 4-substituted-3,4-dihydrobenzo[h]quinoline-2,5,6(1H)-triones as NQO1-directed anti-tumor agents were designed, synthesized, biologically evaluated. Compounds 3n, 3o and 3j proved to be good NQO1 substrates that showed increased metabolic rates relative to that of beta-lapachone. In addition, 3n, 3o and 3j potently inhibited the growth of NQO1-rich breast cancer MCF-7 cell, liver hepatocellular HepG2 cell, and lung cancer A549 cell. In cellular mechanistic studies, the representative compound 3o triggered ROS generation depending on the NQO1 dose, and induce HepG2 cell apoptosis by the generated oxidative stress. Picolinamide group assisted sp(3) C-H bond oxidation of methylene groups to the corresponding carbonyl compounds has been achieved by using simple bottle ferrocene as catalyst and Cu(OAc)(2) or tert-butyl peroxybenzoate (TBPB) as oxidant under mild conditions. This method is applicable for picolinamide bound organic as well as organometallic compounds with yields in the range of 46-82%. Control experiments and mechanistic studies indicate that a radical mechanism is responsible for these oxidative transformations in which ferrocene acts as a catalyst.

Sodium is the most abundant alkali metal in the Earth’s crust and the ocean. However, organosodium compounds have long been considered inferior to organolithium compounds, which have instead dominated synthetic organic chemistry during the last century. Despite being largely neglected because of their reactive nature, it is worth re-exploring organosodium chemistry, in light of the growing demand for sustainable syntheses without recourse to less abundant elements such as lithium. Herein, we demonstrate that, contrary to common belief, organosodium compounds can be easily prepared from aryl chlorides or (hetero) arenes and easy-to-handle sodium dispersion and, after being transmetallated to the corresponding zinc and boron compounds, they readily participate in the Negishi and Suzuki-Miyaura cross-coupling reactions, fundamental carbon-carbon bond-forming reactions in organic synthesis. Direct coupling reactions with organosodium species were also possible.

A trifluoroacetic-acid-mediated desulfurilative sulfonylthiolation of arenes using SS-morpholino dithiosulfonate is described. This system is based on selective activation of the morpholino group over the tosyl group of the doubly transformable sulfur surrogate. Mechanistic studies suggested that the reaction proceeds through electrophilic aromatic substitution followed by sulfur extrusion. The wide substrate scope of this reaction and the transformability of the resulting thiosulfonates enable expeditious access to divergent multifunctionalized sulfides.

Herein, a novel ON-OFF colorimetric and fluorometric chemosensor; 1N-allyl-2-(2, 5-dimethoxyphenyl)-4, 5-diphenyl-1H-imidazole (ADPPI), was constructed for sequential determination of Cu2+ and S2- ions in aqueous media. The interaction between chemosensor ADPPI and different metal cations was investigated using UV-VIS and fluorimetric spectroscopy. ADPPI showed a favorable and good interaction with Cu2+ ions producing blue colored solution peaked at 610 nm with blue fluorescence at lambda(em) = 447 nm. The produced complex between Cu2+ ions and ADPPI can be used as a cascade probe for detection of S2- ions. The detection limits (LODs) were 1.01 nM and 1.25 mu M for Cu2+ and S2- ions, respectively (the lowest between the family of colorimetric and fluorometric chemosensors).
