This article is the first attempt to describe substituent effects in terms of total energies of the substituted derivatives of benzoic acid. The key quantity in this attempt is the difference of the total energies of the para and meta conformers. This quantity is then correlated with Hammett substituent constants and with some theoretical parameters of the electronic structure of the substituent. While it was previously unclear that the total energy of a molecule would prove to be a useful quantity in studying substituent effects, our research shows that, indeed, this energetical difference correlates well with other parameters if only they also relate to both of these conformers, that is they are differences of their individual counterparts determined for both of these conformers separately.

Flavin-based catalysts are photoactive in the visible range which makes them useful in biology and chemistry. Herein, we present electrospray-ionization mass-spectrometry detection of short-lived intermediates in photooxidation of toluene catalysed by flavinium ions (Fl(+)). Previous studies have shown that photoexcited flavins react with aromates by proton-coupled electron transfer (PCET) on the microsecond time scale. For Fl(+), PCET leads to FlH(.+) with the H-atom bound to the N5 position. We show that the reaction continues by coupling between FlH(.+) and hydroperoxy or benzylperoxy radicals at the C4a position of FlH(.+). These results demonstrate that the N5-blocking effect reported for alkylated flavins is also active after PCET in these photocatalytic reactions. Structures of all intermediates were fully characterised by isotopic labelling and by photodissociation spectroscopy. These tools provide a new way to study reaction intermediates in the sub-second time range.

A collection of N-substituted quinolin-2(1H)-ones were screened against a panel of clinically relevant protozoa (Leishmania, Trypanosoma and Acanthamoeba). Three quinolin-2(1H)-one compounds were identified as selective anti-Acanthamoeba agents. Further assessment revealed that these compounds were active against both trophozoite and cyst forms of A. castellanii Neff, and caused protozoa death via apoptosis. The data presented herein identify N-acyl quinolin-2(1H)-ones as a promising new class of selective anti-Acanthamoeba agents.

Cyperus esculentus tubers are the raw material to prepare tigernut milk (horchata), which can be marketed under Protected Designation of Origin (PDO) C hufa de Valencia. The aim of this study was to characterize commercial tigernut milks and compare PDO and non-PDO products. The following aspects were studied: (i) volatile profile, (ii) descriptive sensory profile, (iii) consumer satisfaction degree. The key volatile compounds were limonene, benzaldehyde, linalool and m-methoxyanisole. Principal component analysis indicated a mix of PDO and nonPDO samples in the groups formed. The highest consumer satisfactions were observed for 2 PDO samples. Penalty analysis showed that 80% of non-PDO samples needed improvements, while this percentage drastically decreased to 40% for PDO samples. The online study proved that a lot of people drink horchata, less people know the PDO Chufa de Valencia and even less people consume the protected product consciously. In conclusion, there was not a clear difference among protected and non-protected tigernut milks respect to volatile compounds but there were differences in the degree of consumer preference. So, it is clear that a lack of knowledge regarding the product and its PDO exists and needs attention.

To explore if the catalytic activity of single-metal species in halometallate catalysts can be enhanced with mixed-metal species, aluminum and gallium were simultaneously introduced to prepare a mixed-metal ionic liquid (IL) through a one pot reaction of [HN222]Cl, AlCl3, and GaCl3 in various amounts. The Lewis acidity in Friedel-Crafts acylation and alkylation reactions of these IL compositions were evaluated and all mixed-metal [HN222][xAlCl(3) + (2 – x)GaCl3]Cl ILs exhibited higher catalytic activity than AlCl3, GaCl3, [HN222][Al2Cl7], or [HN222][Ga2Cl7]. The broadening in Al-27 nuclear magnetic resonance (NMR) of these ILs indicates the presence of dynamic anionic species, whereas matrix-assisted laser-desorption ionization time of-flight (MALDI-TOF) and acetonitrile-probed Fourier transform infrared (FT-IR) analyses correlates the structure-activity relationship, for instance, identifying the presence of mixed-metal-containing [Cl2GaOClAlClOGaCl2](-) species in the better-performing ILs.

Stereochemistry of direct conversion of polyamides to hydroxyesters was investigated using model compounds. Optically active secondary-alkyl amines underwent the conversion to corresponding secondary alcohols in moderate yields by treatment with supercritical methanol in the presence of glycolic acid. The reaction progressed through almost completely stereochemical inversion to give secondary alcohols in high yields. Transition-metal-catalyzed sp(2) C-N bond formation is a reliable method for the synthesis of aryl amines. Catalytic sp(3) C-N formation reactions have been reported occasionally, and methods that can realize both sp(2) and sp(3) C-N formation are relatively unexplored. Herein, we address this challenge with a method of catalytic decarboxylative C-N formation that proceeds through a cascade carboxylic acid activation, acyl azide formation, Curtius rearrangement and nucleophilic addition reaction. The reaction uses naturally abundant organic carboxylic acids as carbon sources, readily prepared azidoformates as the nitrogen sources, and 4-dimethylaminopyridine (DMAP) and Cu(OAc)(2) as catalysts with as low as 0.1 mol % loading, providing protected alkyl, alkenyl and aryl amines in high yields with gaseous N-2 and CO2 as the only byproducts. Examples are demonstrated of the late-stage functionalization of natural products and drug molecules, stereospecific synthesis of useful alpha-chiral alkyl amines, and rapid construction of different ureas and primary amines.
