Microglia-mediated neuroinflammation is one of the key mechanisms involved in acute brain injury and chronic neurodegeneration. This study investigated the inhibitory effects of 2-hydroxy-4-methylbenzoic anhydride (HMA), a novel synthetic derivative of HTB (3-hydroxy-4-trifluoromethylbenzoic acid) on neuroinflammation and underlying mechanisms in activated microglia in vitro and an in vivo mouse model of Parkinson’s disease (PD). In vitro studies revealed that HMA significantly inhibited lipopolysaccharide (LPS)-stimulated excessive release of nitric oxide (NO) in a concentration dependent manner. In addition, HMA significantly suppressed both inducible NO synthase and cyclooxygenase-2 (COX-2) at the mRNA and protein levels in LPS-stimulated BV-2 microglia cells. Moreover, HMA significantly inhibited the proinflammatory cytokines such as interleukin (IL)-1beta, IL-6, and tumor necrosis factor-alpha in LPS-stimulated BV-2 microglial cells. Furthermore, mechanistic studies ensured that the potent anti-neuroinflammatory effects of HMA (0.1, 1.0, and 10 mu M) were mediated by phosphorylation of nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha (I kappa B alpha) in LPS-stimulated BV-2 cells. In vivo evaluations revealed that intraperitoneal administration of potent neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 20 mg/kg, four times a 1 day) in mice resulted in activation of microglia in the brain in association with severe behavioral deficits as assessed using a pole test. However, prevention of microglial activation and attenuation of Parkinson’s disease (PD)-like behavioral changes was obtained by oral administration of HMA (30 mg/kg) for 14 days. Considering the overall results, our study showed that HMA exhibited strong anti-neuroinflammatory effects at lower concentrations than its parent compound. Further work is warranted in other animal and genetic models of PD for evaluating the efficacy of HMA to develop a potential therapeutic agent in the treatment of microglia-mediated neuroinflammatory disorders, including PD.

A metal- and oxidant-free electrochemical synthesis of aryl sulfides was developed through a C-H sulfidation reaction of arenes and disulfides. Compared with traditional organic synthesis methods, this direct electrochemical approach efficiently generates aryl sulfides under catalyst- and oxidant-free conditions with the superiorities of wide substrate compatibility, mild reaction condition and waster free. At room temperature, various aryl thiols could be transformed smoothly in an undivided cell. Based on cyclic voltammetry (CV) and control experiments, the possible reaction mechanism was also proposed. The gram-scale synthesis emphasizes the practicability of this electrochemical strategy. Cyperus esculentus tubers are the raw material to prepare tigernut milk (horchata), which can be marketed under Protected Designation of Origin (PDO) C hufa de Valencia. The aim of this study was to characterize commercial tigernut milks and compare PDO and non-PDO products. The following aspects were studied: (i) volatile profile, (ii) descriptive sensory profile, (iii) consumer satisfaction degree. The key volatile compounds were limonene, benzaldehyde, linalool and m-methoxyanisole. Principal component analysis indicated a mix of PDO and nonPDO samples in the groups formed. The highest consumer satisfactions were observed for 2 PDO samples. Penalty analysis showed that 80% of non-PDO samples needed improvements, while this percentage drastically decreased to 40% for PDO samples. The online study proved that a lot of people drink horchata, less people know the PDO Chufa de Valencia and even less people consume the protected product consciously. In conclusion, there was not a clear difference among protected and non-protected tigernut milks respect to volatile compounds but there were differences in the degree of consumer preference. So, it is clear that a lack of knowledge regarding the product and its PDO exists and needs attention.

A new protocol for the chemoselective aromatization of tetrahydro-beta-carboline-3-carboxylic acids to direct biomimetic one step synthesis of beta-carboline-3-carboxylic acids 2 A-L using catalytic amount of I-2 in DMSO/H+ produces very good yield. This method was also successfully extended for the aromatization of tetrahydro-beta-carboline-3-methyl esters 5 A-G as well as one-step synthesis of Marinacarbolines-D analog, respectively. Further, the mechanism and role of iodine-DMSO in aromatization has been studied by Density Functional Theory Calculation.

Mn doped zirconia is utilized as an environmental-friendly and efficient catalyst for an ultrasound mediated four-component coupling reaction, containing dimethylacetylenedicarboxy late/ethyl acetoacetate, hydrazine hydrate, malononitrile, and aromatic aldehyde. These reactions were performed under green solvent conditions, to yield pyrano[2,3-c]pyrazole-3-carboxylate/pyr ano[2,3-c]pyrazole-5-carbonitrile derivatives (5a-g and 7a-g) with good to excellent yields (88-98%). The structures of the compounds were identified and confirmed by H-1 NMR, N-15 NMR, C-13 NMR, FT-IR and HR-MS spectral data. The prepared catalyst Mn/ZrO2 was synthesized and fully characterized by various techniques including P-XRD, BET, SEM and TEM analysis. The main benefits of this process are short reaction times, easy work-up, reusability of the catalyst and no chromatographic purifications. (C) 2016 The Authors. The degradation of organic contaminants in aquatic systems has raised immense attention worldwide, and the second-order rate constant (k(SO4)(-)) of water pollutants oxidized by sulfate radical anion is an important index for assessing the degradation efficiency of organics. Herein, a new norm mathematical formula is defined. Based on this, four new descriptors are proposed and a QSPR model is developed for predicting k(SO4)(-) using 30 families of emerging organic pollutants in water. The statistical results fully prove that this model has good fitting effect and stability with R-2 of 0.8862, Q(LOO)(2) of 0.8466, and Q(5-fold)(2) of 0.8329, respectively. The validation results including cross validation, applicability domain analysis, and model comparison show that this model has good robustness, predictive performance, and reliability. These decent results indicate that the new norm mathematical formula is effective in calculating descriptors and the norm indexes have a great application for evaluating the transformation fate of organic pollutants by sulfate radical in aquatic systems.

High-mannose (Man(9)GlcNAc(2)) is the main carbohydrate unit present in viral envelope glycoproteins such as gp120 of HIV and the GP1 of Ebola virus. This oligosaccharide comprises the Man(9)epitope conjugated to two terminalN-acetylglucosamines by otherwise rarely-encountered beta-mannose glycosidic bond. Formation of this challenging linkage is the bottleneck of the few synthetic approaches described to prepare high mannose. Herein, we report the synthesis of the Man(9)epitope with both alpha and beta configurations at the reducing end, and subsequent evaluation of the impact of this configuration on binding to natural receptor of high-mannose, DC-SIGN. Using fluorescence polarization assays, we demonstrate that both anomers bind to DC-SIGN with comparable affinity. These relevant results therefore indicate that the more synthetically-accesible Man(9)alpha epitope may be deployed as ligand for DC-SIGN in bothin vitroandin vivobiological assays.

Experimental solubilities have been determined for anthracene, benzil, 2-chloroanthraquinone, 9-fluorenone, 2-hydroxybenzoic acid, 2-methoxybenzoic acid, 2-methylbenzoic acid, 3-methylbenzoic acid, phenothiazine, pyrene, and thioxanthen-9-one dissolved in benzyl alcohol at 298.15 K. The measured solubility data, combined with previously published activity coefficient and solubility data, are used to determine Abraham model correlations for solute transfer to benzyl alcohol from both water and from the gas phase. The derived Abraham model correlations were found to back-calculate the experimental partition coefficients and solubility ratios to within 0.14 log units (or less).

Diflubenzuron (DFB) and fenuron (FEN) are benzoylurea and phenylurea pesticides, widely used in Senegal, that do not exhibit any natural fluorescence, but can be determined by means of photoinduced fluorescence (PIF) methods. Photodegradation of DFB and FEN yielded a number of fluorescent and non-fluorescent photoproducts. For both pesticides, at least 10 photoproducts were detected and identified by gas chromatography-mass spectrometry (GC/MS).
