Herein, we present the first report on TiO2 NPs based hybrid nano-reactor designed and synthesized via N-(2-amino ethyl)-3-amino propyl trimethoxy silane (AAPTMS) functionalization of graphene oxide (GO). The assynthesized nano-reactor was characterized by means of several analytical-cum-spectroscopic techniques that support the successful attempt for the synthesis of TiO2@AAPTMS-rGO. Further, the present catalyst was evaluated for its catalytic performance towards an expedient access to biologically relevant synthesis of 2,4,5-triaryl imidazoles. The functional sites of TiO2@AAPTMS-rGO direct the catalysis from the reactant to product with high atom economy, shorter reaction time and excellent yields. Moreover, the present catalyst TiO2@AAPTMS-rGO shows promising reusability for several catalytic cycles. It also provides a robust approach for 2,4,5-triaryl imidazoles with improved performance and without significant loss in catalytic activity. Further, this catalyst can be upgraded for other organic transformations leading to numerous pharmaceuticals and environment benefits.

Fluorine-containing compounds have been widely used in the fields of pharmaceuticals, agrochemicals and functional materials, mainly due to the well-known fluorine effect of the fluoroalkyl groups on the physical, chemical and biological properties of molecules. Tri- and difluoromethyl ethers play an important role in many medicinally compounds. Among various fluorinated moieties, ORf-containing groups have attracted much more attention very recently owing to the impressive conformational changes and maximal shifts in electron distribution brought by fluorine. The alpha-fluorine substitution of ethers shortens and strengthens the C-O bond and thus improves the in vivo oxidative stability of the ether moiety of a drug. Over the past few decades, there are some reliable ways on accessing trifluoromethyl ethers and difluomethyl ethers. Considering the importance of synthesis of monofluoromethoxy arenes and the substrate limitation (phenols or alcohols) of current state, a method was developed to access monofluoromethoxy arenes from aryl halides, arylboronic acids and arenes via a one-pot synthesis. Phenols can be prepared by the hydroxylation of aryl halides catalyzed by transition-metal complexes. In this work, a new strategy was envisioned a two-step sequence for the conversion of aryl halides to monofluoromethoxy arenes based on the palladium-catalyzed conversion of aryl phenols and in situ conversion of the resulting phenoxides with monofluoromethylating reagents. The investigation began with optimization of the conversion of 1-chloro-4-methoxy-benzene. The approach was achieved by using Pd-2(dba)(3) (2 mol%) as the catalyst under an inert atmosphere, di-tertbuty1(2′,4′,6′-triisopropyl-[1,1′-biphenyl]-2-yl)phosphane (8 mol%) as the ligand, KOH (1 equiv.) as the nucleophile, and 1,4-dioxane/H2O (V : V=5 : 3) as the solvent. Further monofluoromethylation used fluoromethyl iodide (2 equiv.) as the monofluoromethylating reagent and CH3CN as the co-solvent. Finally, the desired product was obtained in 82% yield. Therefore, this method was also applied to drugs, for example, Loratadine could be converted to the corresponding product (2o) in 53% yield and Fenofibrate, reacting to form the monofluoromethoxy arenes (2p) in modest yield. We report the practical synthesis of a key fragment of islatravir (MK-8591), a novel nucleoside reverse transcriptase translocation inhibitor (NRTTI) currently under investigation for treatment and pre-exposure prophylaxis (PrEP) against HIV infection. The fragment, the unnatural nucleobase 2-fluoroadenine, is incorporated into MK-8591 via a biocatalytic aldolglycosylation cascade, which imposes stringent requirements for its synthesis and isolation. Presented herein is the development work leading to a practical, scalable route from guanine, featuring a dual fluorination approach to a novel 9-THP-2,6-difluoropurine intermediate that enables a mild, highly selective, direct amination. This one-pot fluorination/amination sequence utilizes a direct isolation to deliver high purity 9-THP-2-fluoroadenine, which features ideal properties with respect to reactivity, solubility, and crystallinity. An acid-catalyzed liberation of 2-fluoroadenine in aqueous buffer delivers the appropriate purity profile to facilitate the enzymatic cascade to access MK-8591.

A series of substituted benzoate-bridged dichromium(II, II) complexes [Cr-2(RCO2)(4)(THF)(2)] ([Cr-2]), where RCO2- is substituted benzoate, was synthesized and its structural and magnetic properties were investigated. Systematic investigations of a Pd(II)-catalyzed, 8-aminoquinoline directing group (DG)-aided sp(2) delta-C-H amidation (C-N bond formation) of different biaryl carboxamides are reported. Various biaryl carboxamides with suitably positioned sp(2) delta-C-H bond with respect to the DG were assembled via beta-C-H arylation and then they were subjected to Pd(II)-catalyzed sp(2) delta-C-H intramolecular amidation/annulation reactions. While the intramolecular amidation of the sp(2) delta-C-H bond of some carboxamides was not fruitful, several biaryl carboxamides underwent intramolecular amidation of their sp(2) delta-C-H bonds to afford various tricyclic quinolone motifs such as, phenanthridin-6(5H)-ones and thieno-/furo-/pyrrolo-[2,3-c]quinolin-4(5H)-ones. The assembly of the required biaryl carboxamides possessing the sp(2) delta-C-H bond via the beta-C-H arylation and the successive intramolecular amidation (C-N bond formation) of the resulting biaryl carboxamides were also performed in one-pot reaction conditions to afford tricyclic quinolones.

Herein, beta-carboline derivative 17c was designed and synthesized with obvious myocardial protective activity for the first time. Pretreatment of 17c effectively protected the cardiomyocyte H9c2 cells from H2O2-induced lactate dehydrogenase leakage and restored the endogenous antioxidants, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px). Besides, 17c effectively protected the mitochondria through decreasing the reactive oxygen species overproduction and enhancing the mitochondrial membrane potential. As a result, 17c significantly reduced the necrosis of cardiomyocytes in H2O2-induced oxidative stress, which was more potent than polydatin. In MI/R injury rats, 17c pretreatment obviously increased the levels of SOD and GSH-Px and inhibited the apoptosis of cardiomyocytes. Through this way, the size of myocardial infarction was significantly reduced after MI/R injury in vivo, better than that of polydatin, suggesting that 17c is a promising cardioprotectant for the prevention of MI/R injury.
