Described herein is a novel one-pot, three-component reaction where aldehydes, electron-rich arenes, and TMSCN in the presence of BF3-OEt2 allowed direct access to a number of diarylacetonitriles under mild reaction conditions in good to excellent yields. Implementation of this assembly protocol to a concise synthetic approach to shoreaphenol, a bioactive oligostilbenoid natural product, is also demonstrated.

This study aimed to elucidate the intrinsic mechanisms of PS activation by carbon nanotubes (CNTs). Singlet oxygen generation (O-1(2)) and direct CNTs-mediated electron transfer were hypothesized to be two major pathways of the oxidation of 2,4-dichlorophenol (2,4-DCP) by PS in the presence of both unmodified and modified CNTs. For the first time, roles of CNT active sites responsible for PS activation were determined using CNT derivatization and structural characterization. By selectively deactivating the carbonyl, hydroxyl or carboxylic groups on CNTs surface and linear sweep voltammetry (LSV) analysis, -C=0 groups were determined to be the main active sites contributing to the direct electron transfer oxidation, while singlet oxygen was generated at CNTs defects. Subsequent UV irradiation was shown to cause the recovery of surface defects with I-D/I-G of CNTs increasing by 21%. This resulted in the regeneration of the performance for the coupled system and allowed for multi-cycle activation of PS by CNTs. These results suggest that CNTs/PS system combined with regeneration based on UV irradiation can be used as an effective alternative process for continuous degradation of recalcitrant aqueous contaminants through the non-radical mechanism. The degradation of organic contaminants in aquatic systems has raised immense attention worldwide, and the second-order rate constant (k(SO4)(-)) of water pollutants oxidized by sulfate radical anion is an important index for assessing the degradation efficiency of organics. Herein, a new norm mathematical formula is defined. Based on this, four new descriptors are proposed and a QSPR model is developed for predicting k(SO4)(-) using 30 families of emerging organic pollutants in water. The statistical results fully prove that this model has good fitting effect and stability with R-2 of 0.8862, Q(LOO)(2) of 0.8466, and Q(5-fold)(2) of 0.8329, respectively. The validation results including cross validation, applicability domain analysis, and model comparison show that this model has good robustness, predictive performance, and reliability. These decent results indicate that the new norm mathematical formula is effective in calculating descriptors and the norm indexes have a great application for evaluating the transformation fate of organic pollutants by sulfate radical in aquatic systems.

Tankyrases 1 and 2 are central biotargets in the WNT/beta-catenin signaling and Hippo signaling pathways. We have previously developed tankyrase inhibitors bearing a 1,2,4-triazole moiety and binding predominantly to the adenosine binding site of the tankyrase catalytic domain. Here we describe a systematic structure-guided lead optimization approach of these tankyrase inhibitors. The central 1,2,4-triazole template and trans-cyclobutyl linker of the lead compound 1 were left unchanged, while side-group East, West, and South moieties were altered by introducing different building blocks defined as point mutations. The systematic study provided a novel series of compounds reaching picomolar IC50 inhibition in WNT/beta-catenin signaling cellular reporter assay. The novel optimized lead 13 resolves previous atropisomerism, solubility, and Caco-2 efflux liabilities. 13 shows a favorable ADME profile, including improved Caco-2 permeability and oral bioavailability in mice, and exhibits antiproliferative efficacy in the colon cancer cell line COLO 320DM in vitro.

Enantioselective electrophilic three-component thioarylation of alkenes by chiral selenide catalysis with free phenols as arylating sources is disclosed. A variety of chiral phenols were achieved in high regio-, enantio-, and diastereoselectivities. Mechanistic studies revealed that this transformation went through carbon nucleophilic attack to give the products rather than the process of intramolecular rearrangement of phenolic ether intermediates. The application of this organocatalytic method in the alkylation of methoxy-substituted benzenes elucidated its generality.

This study presents an efficient synthesis of 3-[6-(substituted-phenyl)-[1,2,4]triazolo[3,4-b][1,3,4] thiadiazol-3-yl]-1H-indazole via dehydrative condensation with cyclization of 4-amino-5-(1H-indazol-3-yl)-4H-[1,2,4]triazole-3-thiol and fluorinated or nonfluorinated carboxylic acids in presence of phosphorous oxychloride. The multistep reaction pathway proceeds through different compounds. Present synthesis has the advantages of easily accessible starting materials, convenient synthesis, simple reaction condition, wider substrate scope, and higher yield (75% to 90% isolated).

In the presence of a [Cp*RhCl2](2) catalyst, the Lewis acid In(OTf)(3), and the mild base Na2CO3, aromatic carboxylates and ,-unsaturated ketones undergo a unique hydroarylation/Claisen/retro-Claisen process to give the corresponding indanones. In this carboxylate-directed ortho-C-H annelation, the C-COR bond of the ketone and the CO-OH group of the aromatic carboxylate are cleaved, and the hydroxy group is transferred from the aromatic to the aliphatic acyl residue. This reactivity is synthetically useful, particularly when starting from cyclic ketones, which are converted into indanones bearing aliphatic carboxylate side chains, thus greatly increasing the molecular complexity of aromatic carboxylates in a single step.
