The structure-activity relationship of a 4-Azaindole-2-piperidine compound selected from GlaxoSmithKline’s recently disclosed open-resource Chagas box and possessing moderate activity against Trypanosoma cruzi, the parasite responsible for Chagas disease, is presented. Despite considerable medicinal chemistry efforts, a suitably potent and metabolically stable compound could not be identified to advance the series into in vivo studies. This research should be of interest to those in the area of neglected diseases and in particular anti-kinetoplastid drug discovery.

A catalyst- and metal-free electrochemical hydrodehalogenation of aryl halides is disclosed. Our reaction by a flexible protocol is operated in an undivided cell equipped with an inexpensive graphite rod anode and cathode. Trialkylamines nBu(3)N/Et3N behave as effective reductants and hydrogen atom donors for this electrochemical reductive reaction. Various aryl and heteroaryl bromides worked effectively. The typically less reactive aryl chlorides and fluorides can also be smoothly converted. The utility of our method is demonstrated by detoxification of harmful pesticides and hydrodebromination of a dibrominated biphenyl (analogues of flame-retardants) in gram scale.

alpha-Aryl oxindoles are accessed from isatin via a two-step procedure involving a phospha-Brook rearrangement and a Friedel-Crafts alkylation in a one-pot procedure. The use of 1,1,1,3,3,3-hexafluoro-2-propanol as solvent significantly extended the reaction substrate scope to include relatively less electron-rich arenes including benzene. This new alkylation method is fast and straightforward and allows for the direct introduction of the oxindole moiety onto a range of aromatic compounds including phenols. Additionally, the application of arylated products was shown in decarboxylative asymmetric allylation and protonation.

Oxidation of organic substrates is achieved in nature under mild conditions thanks to metalloenzymes but remains a challenge for chemists. Herein we show by UV-Vis spectroelectrochemistry that when (MnTPPCl)-T-III is electrochemically reduced to Mn-II in CH2Cl2 under O-2, a (MnO2)-O-II species is generated. Benzoic anhydride reacts with the latter triggering a catalytic current in cyclic voltammetry. Electrolysis on the catalytic wave in the presence of cyclooctene leads to its oxygenation or halogenation depending on the axial ligand present as reported here for the first time.

Fluorine-containing compounds have been widely used in the fields of pharmaceuticals, agrochemicals and functional materials, mainly due to the well-known fluorine effect of the fluoroalkyl groups on the physical, chemical and biological properties of molecules. Tri- and difluoromethyl ethers play an important role in many medicinally compounds. Among various fluorinated moieties, ORf-containing groups have attracted much more attention very recently owing to the impressive conformational changes and maximal shifts in electron distribution brought by fluorine. The alpha-fluorine substitution of ethers shortens and strengthens the C-O bond and thus improves the in vivo oxidative stability of the ether moiety of a drug. Over the past few decades, there are some reliable ways on accessing trifluoromethyl ethers and difluomethyl ethers. Considering the importance of synthesis of monofluoromethoxy arenes and the substrate limitation (phenols or alcohols) of current state, a method was developed to access monofluoromethoxy arenes from aryl halides, arylboronic acids and arenes via a one-pot synthesis. Phenols can be prepared by the hydroxylation of aryl halides catalyzed by transition-metal complexes. In this work, a new strategy was envisioned a two-step sequence for the conversion of aryl halides to monofluoromethoxy arenes based on the palladium-catalyzed conversion of aryl phenols and in situ conversion of the resulting phenoxides with monofluoromethylating reagents. The investigation began with optimization of the conversion of 1-chloro-4-methoxy-benzene. The approach was achieved by using Pd-2(dba)(3) (2 mol%) as the catalyst under an inert atmosphere, di-tertbuty1(2′,4′,6′-triisopropyl-[1,1′-biphenyl]-2-yl)phosphane (8 mol%) as the ligand, KOH (1 equiv.) as the nucleophile, and 1,4-dioxane/H2O (V : V=5 : 3) as the solvent. Further monofluoromethylation used fluoromethyl iodide (2 equiv.) as the monofluoromethylating reagent and CH3CN as the co-solvent. Finally, the desired product was obtained in 82% yield. Therefore, this method was also applied to drugs, for example, Loratadine could be converted to the corresponding product (2o) in 53% yield and Fenofibrate, reacting to form the monofluoromethoxy arenes (2p) in modest yield. One-pot method to access aryl monofluoromethyl ethers from arylboronic acids and arenes were also under consideration and the yields were objective.

We report the practical synthesis of a key fragment of islatravir (MK-8591), a novel nucleoside reverse transcriptase translocation inhibitor (NRTTI) currently under investigation for treatment and pre-exposure prophylaxis (PrEP) against HIV infection. The fragment, the unnatural nucleobase 2-fluoroadenine, is incorporated into MK-8591 via a biocatalytic aldolglycosylation cascade, which imposes stringent requirements for its synthesis and isolation. Presented herein is the development work leading to a practical, scalable route from guanine, featuring a dual fluorination approach to a novel 9-THP-2,6-difluoropurine intermediate that enables a mild, highly selective, direct amination. This one-pot fluorination/amination sequence utilizes a direct isolation to deliver high purity 9-THP-2-fluoroadenine, which features ideal properties with respect to reactivity, solubility, and crystallinity. An acid-catalyzed liberation of 2-fluoroadenine in aqueous buffer delivers the appropriate purity profile to facilitate the enzymatic cascade to access MK-8591.

Highly selective direct monofluorination of indoles and arenes was developed through an approach that allows site-specific solubility of substrate and fluorine source in the micelle. This approach was highly selective for a broad range of substrates with excellent functional group tolerance. Differences in binding constant and solubility of indoles and arenes in the micelle allowed the fine-tuning of selectivity. Control experiments suggested a radical pathway and provided insight into the role of micelles of the environmentally benign amphiphile PS-750-M. Dynamic light scattering experiments strongly indicated the site-specific solubility of the substrate and fluorine source. The methodology was successfully adapted to gram scale, and the E-factor established from a recycle study indicated that the process is environmentally responsible and sustainable.

Well-defined ruthenium(II) biscarboxylate complexes enabled selective ortho-deuteration with weakly-coordinating, synthetically useful carboxylic acid with outstanding levels of isotopic labeling.
