The alpha 7 nicotinic acetylcholine receptor (alpha 7 nAChR) is a ligand-gated ion channel that is involved in cognition disorders, schizophrenia, pain, and inflammation. Allosteric modulation of this receptor might be advantageous to reduce the toxicity in comparison with full agonists. Our previous results obtained with some hydroxy-chalcones, which were identified as positive allosteric modulators (PAMs) of alpha 7 nAChR, prompted us to evaluate the potential of some structurally related naturally occurring flavonoids and curcuminoids and some synthetic curcumin analogues, with the aim of identifying new allosteric modulators of the alpha 7 nAChR. Biological evaluation showed that phloretin, demethoxycurcumin, and bis-demethoxicurcuming behave as PAMs of alpha 7 nAChR. In addition, some new curcumin derivatives were able to enhance the signal evoked by ACh; the activity values found for the tetrahydrocurcuminoid analog 23 were especially promising.

A ligand-promoted palladium (II)-catalyzed synthesis of arylalkynes and phthalides from benzoic acids and bromoalkynes via carboxylate-assisted ortho-C-H activation is reported. A series of phthalides with various functional groups are prepared via ortho-alkynylation and alkynylation-annulation. Moreover, the key ortho-alkynylated products are also obtained by controlling the reaction conditions. In addition, heteroaryl acids could react smoothly to form the corresponding alkynylation and cyclization products.

The highly enantioselective copper-catalyzed three-component boroacylation of 1,1-disubstituted allenes is reported by using a class of chiral ligands (WJ-Phos), delivering various functionalized organoboron compounds bearing an all-carbon stereocenter in moderate to good yields with high enantioselectivities. WJ-Phos is a ferrocene-derived chiral sulfinamide phosphine ligand and can be easily synthesized in gram-scale from readily available starting materials in short steps. The salient features of this reaction include moderate to good yields, high enantioselectivities, gram-scale synthesis, diverse synthetic transformations, and the development of a new chiral ligand.

A multi-gram-scale synthetic route to a novel, bifunctional normorphan 3D building block bearing orthogonally reactive vinyl MIDA boronate and N-2,4-dimethoxybenzyl (DMB) amide functional handles has been developed. Synthetic manipulation at each of the functional handles has been demonstrated including Suzuki-Miyaura cross-coupling, exo-diastereoselective hydrogenation, N-DMB group removal and lactam reduction. Normorphan cores derived from the building block satisfied AstraZeneca’s ‘rule of 2’ for building blocks and had a high fraction of sp(3) hybridised carbon atoms (Fsp(3)). A virtual lead-like library of 344 compounds derived from the building block had attractive lead-like properties. Microplastics (MPs) and nanoplastics (NPs) are ubiquitous and contaminate soil, surface waters, atmospheric aerosol, precipitations, indoor and outdoor environments. However, the occurrence, transformation and fate of NPs in the environment are still unclear. In this work, polystyrene nanoparticles (PS-NPs) are used as a proxy of NPs to study their reactivity and potential impact on atmospheric and surface waters. In particular, the reactivity with hydroxyl radicals (center dot OH) in the aqueous phase is investigated. For the first time, a reactivity constant for the reaction of NPs with center dot OH is measured, strongly dependent on the exposed partide surface area of NPs. Degradation products (short chain carboxylic acids and aromatic compounds), obtained by direct and center dot OH-mediated photolysis of PS-NPs suspensions, are identified by mass spectrometry. Irradiation of a PS-NPs suspension under natural sunlight for 1 year has shown the formation of formic acid and organic compounds similar to those found in riverine and cloud dissolved organic matter, which could contribute significantly to the dissolved organic matter in the aqueous phase. A simple, facile and efficient method has been developed for the Friedel-Crafts benzoylation of arenes using magnetic gamma-Fe2O3 nanoparticles under solvent-free sonication. The gamma-Fe2O3 nanoparticles were used as an efficient and magnetically recoverable catalyst for the synthesis of aromatic ketones in good to excellent yields at room temperature under solvent-free. The reaction occurred with high regioselectivity under mild condition. The magnetic gamma-Fe2O3 nanoparticles are economically synthesized in large-scale, easily separated from the reaction mixture by an external magnet and able to be reused several times without significant loss of the catalytic performance, which make them easy application to industrial processes. A diverse range of ureas, N-acylureas, carbamates, and thiocarbamates has been synthesized in good to excellent yields by reacting N-acylbenzotriazoles individually with amines or amides or phenols or thiols in the presence of diphenylphosphoryl azide (DPPA) as a suitable azide donor in anhydrous toluene at 110 degrees C for 3-4 hours. In this route, DPPA was found to be a good alternative to trimethylsilyl azide and sodium azide for the azide donor in Curtius degradation. The high reaction yields, one-pot and metal-free conditions, straightforward nature, easy handling, use of readily available reagents, and in many cases avoidance of column chromatography are the notable features of the devised protocol.

The chlorination of a bioactive compound can change its physiological properties and improve its pharmacokinetic and pharmacological profiles. It therefore has been an important strategy for drug discovery and development. However, the direct aromatic chlorination of complex bioactive molecules is too difficult to be practical. In fact, many functional groups such as hydroxyls, amines, amides or carboxylic acids may strongly restrain the reactivity of Cl+ by forming a halogen bond. Here we report a highly efficient aromatic chlorination of arenes that is catalysed by dimethyl sulfoxide with N-chlorosuccinimide as the chloro source. The mild conditions, easy-availability and stability of the catalyst and reagents, as well as good functional-group tolerance, showed the approach to be a versatile protocol for the late-stage aromatic chlorination of complex natural products, drugs and peptides. The multi-gram experiment and low-cost of N-chlorosuccinimide and dimethyl sulfoxide shows great potential for drug discovery and development in industrial applications. Late-stage aromatic chlorination of active pharmaceutical ingredients has enormous potential in drug discovery yet still features limited applicability due to issues of functional-group tolerance. Now, dimethyl sulfoxide is reported as catalyst for the chlorination of a diverse family of bioactive molecules in combination with N-chlorosuccinimide.
