The Rh-catalyzed C-H alkylation of benzylamine derivatives with unactivated 1-alkenes that proceeds via a picolinamide directing group is reported. The crucial role of an acid additive in this transformation is confirmed. Aromatic acids showed high linear selectivity, and aliphatic acids provided branched alkylation products as the major product. The reaction has a broad scope for benzylamines and alkenes. Deuterium labeling experiments suggest that a Rh-carbene intermediate is involved in the case of linear product formation. A different reaction pathway, however, appears to be involved in the case of branched alkylation products, and this pathway also appeared to be a minor pathway in linear-selective reactions.

A new series of quinazolinone derivatives bearing pyridine, pyrimidine, pyrazole, or pyran moieties were synthesized for the purpose of anticancer cell line evaluation. Synthesis of these derivatives was achieved by the reaction of the ketone 2 with the appropriate aldehydes in the presence of either ethylcyanoacetate or malononitrile and ammonium acetate in one-pot reaction. Chalcones 6 reacted also with hydrazine hydrate to give the corresponding pyrazolines 7 and reacted with urea or thiourea to give the 2-oxopyrimidines or the 2-thioxopyrimidines 8, respectively. Evaluation of some representative examples of the newly synthesized compounds against cancer cell lines showed promising activity as anticancer agents.

Preparation of an ordered mesoporous polypyrrole/carbon (PPy/OMC) composite has been described through a two-step nanocasting process using KIT-6 as a template. Characterization of the PPy/OMC nanocomposite by various analysis methods such as TEM, XRD, TGA, SEM and N-2 sorption confirmed the preparation of a material with ordered mesoporous structure, uniform pore size distribution, high surface area and high stability. This nanocomposite was then used for the immobilization of palladium nanoparticles. The nanoparticles were almost uniformly distributed on the support with a narrow particle size of 20-25 nm, confirmed by various analysis methods. Performance of the Pd@PPy/OMC catalyst was evaluated in the aerobic oxidation of various primary and secondary alcohols on water as a green solvent, giving the corresponding carboxylic acids and ketones in high yields and excellent selectivity. The catalyst could also be reused for at least 10 reaction runs without losing its catalytic activity and selectivity. High catalytic efficiency of the catalyst can be attributed to a strong synergism between the PPy/OMC and that of supported Pd nanoparticles.

Here, we explore the GO-catalyzed cross-dehydrogenative coupling of oxindoles with arenes and thiophenols for the rapid synthesis of 3-aryloxindoles and 3-sulfenylated oxindoles. Control experiments and small-molecule mimicking studies reveal that the acidic nature and quinone-type functionalities of GO are synergistically utilized for the coupling reaction. The reaction proceeds under simple and mild reaction conditions, exhibits good functional group tolerance, and can be easily scaled up to the gram level.

This study reported a stereoselective synthesis of (+)/(+)-pentenomycin I in 4-5 steps through regioselective silylation, optical resolution and dihydroxylation, followed by an olefin formation, from a known racemic cyclopentenone prepared from 2-deoxy-o-glucose. We also accomplished the transformation of a common intermediate into a variety of analogs. In addition, the antimicrobial activities of the pentenomycin analogs were evaluated, which revealed important structural factors of pentenomycins for the antimicrobial activities. Solvent reorganization is a major driving force of protein-ligand association, but the contribution of binding site waters to ligand affinity is poorly understood. We investigated how altered interactions with a water network can influence ligand binding to a receptor. A series of ligands of the A(2A) adenosine receptor, which either interacted with or displaced an ordered binding site water, were studied experimentally and by molecular dynamics simulations. An analog of the endogenous ligand that was unable to hydrogen bond to the ordered water lost affinity and this activity cliff was captured by molecular dynamics simulations. Two compounds designed to displace the ordered water from the binding site were then synthesized and evaluated experimentally, leading to the discovery of an A(2A) agonist with nanomolar activity. Calculation of the thermodynamic profiles resulting from introducing substituents that interacted with or displaced the ordered water showed that the gain of binding affinity was enthalpy driven. Detailed analysis of the energetics and binding site hydration networks revealed that the enthalpy change was governed by contributions that are commonly neglected in structure-based drug optimization. In particular, simulations suggested that displacement of water from a binding site to the bulk solvent can lead to large energy contributions. Our findings provide insights into the molecular driving forces of protein-ligand binding and strategies for rational drug design.

A polystyrene-immobilized isothiourea has been applied to the enantioselective acylative kinetic resolution (KR) of monoacylated BINOL(s) with inexpensive isobutyric anhydride in batch and flow. High selectivity values (s=29 at 0 degrees C) and a remarkable stability of the catalytic system in the operation conditions have been recorded for unsubstituted BINOL. No significant loss of activity/selectivity is recorded after 10 consecutive KR cycles in batch. A continuous flow process has been implemented and operated with a 100 mmol (32.8 g) sample of racemic monoacetylated BINOL in dichloromethane solution in an 84 hours experiment with a packed bed reactor containing 1 g (f=0.37 mmol.g(-1)) of the functional resin (s=17-21). Residence time can be decreased to 10 min with the same reactor to achieve a conversion of 58% with a selectivity factor s=17 when a more highly functionalized catalyst (f=0.88 mmol.g(-1)) is used. This translates into a remarkable combined productivity of 5.5 mmol(prod) . mmol(cat)(-1) . h(-1).

Supercritical fluid chromatography is becoming more prevalent, particularly in industry. This is due to the inexpensive, and more importantly, environmentally benign carbon dioxide that is used as the major component of the mobile phase. Water is minimally miscible with carbon dioxide at temperatures and pressures commonly used in SFC. However, the introduction of a polar alcohol modifier component increases the solubility of water in carbon dioxide. Previously, the addition of small amounts of water in the mobile phase was shown to provide significant gains in efficiency in chiral supercritical fluid chromatography, especially with polar stationary phases. In this work, we report the effect of the addition of small amounts of water on efficiency and retention factor with four different SFC stationary phases used for achiral analysis namely FructoShell-N (native cyclofructan-6), SilicaShell (bare silica), PoroShell 120 EC C18 (octadecyl silica) and Xselect C18 SB. This is the first reported use of FructoShell-N, a cyclofructan derivatized phase for SFC applications. We devised a predictive test to determine which analytes show an increase in efficiency using their known chemical constants (logK(ow), pK(a), PSA and H-sum). We also use discriminant analysis to elucidate the most important analyte parameters that contribute to water enhanced efficiency gains. (C) 2020 Elsevier B.V. All rights reserved.

2-Pyridyl ketones widely appear in bioactive molecules, natural products, and are employed as precursors of chiral 2-pyridine alky/aryl alcohols or 2-aminoalkyl pyridine ligands for asymmetric catalysis. Herein, a practical method for the rapid synthesis of 2-pyridyl ketone library in continuous flow is reported, in which the 2-lithiopyridine formed by Br/Li exchange reacts with commercially available esters to obtain 2-pyridyl ketones in a good yield at short reaction time. This protocol functions broadly on a variety of esters and has been applied to the synthesis of TGF-beta type 1 receptor inhibitor LY580276 intermediate in an environmentally friendly method. It is rapid, reliable, and cost-efficient to afford diverse kinds of 2-pyridyl ketones in the compound library.
