Evidence for a novel binding site conformer of aldose reductase in ligand-bound state

Human aldose reductase (ALR2) has evolved as a promising therapeutic target for the treatment of diabetic long-term complications. The binding site of this enzyme possesses two main subpockets: the catalytic anion-binding site and the hydrophobic specificity pocket. The latter can be observed in the open or closed state, depending on the bound ligand. Thus, it exhibits a pronounced capability for induced-fit adaptations, whereas the, catalytic pocket exhibits rigid properties throughout all known crystal, structures. Here, we determined two ALR2 crystal structures at 1.55 and 1.65 angstrom resolution, each complexed with an inhibitor of the recently described naphtho[1,2-d]isothiazole acetic acid series. In contrast to the original design hypothesis based on the binding mode of tolrestat (1), both inhibitors leave the specificity pocket in the closed state. Unexpectedly, the more potent ligand (2) extends the catalytic pocket by opening a novel subpocket. Access to this novel subpocket is mainly attributed to the rotation of an indole moiety of Trp 20 by about 35 degrees. The newly formed subpocket provides accommodation of the naphthyl portion of the ligand. The second inhibitor, 3, differs from 2 only by an extended glycolic ester functionality added to one of its carboxylic groups. However, despite this slight structural modification, the binding mode of 3 differs dramatically from that of the first inhibitor, but provokes less pronounced induced-fit adaptations of the binding cavity. Thus, a novel binding site conformation has been identified in a region where previous complex structures suggested only low adaptability of the binding pocket. Furthermore, the two ligand complexes represent an impressive example of how the slight change of a chemically extended side-chain at a given ligand scaffold can result in a dramatically altered binding mode. Thermolysis of 1,4,2-dithiazol-5-ones in the presence of dimethyl acetylenedicarboxylate yielded dimethyl isothiazole-4,5-dicarboxylates, together with sulfur, carbon oxysulfide and nitriles. The proposed mechanism involves initial expulsion of carbon oxysulfide, followed by 1,3-dipolar cycloaddition of the resulting nitrile sulfide to the alkyne. Synthesis of Isothiazole via the Rhodium-Catalyzed Transannulation of 1,2,3-Thiadiazoles with Nitriles

A synthetic method for obtaining a wide variety of isothiazoles by the Rh-catalyzed transannulation of 1,2,3-thiadiazoles with alkyl, aryl, and heteroaryl nitriles, which proceeds via an alpha-thiavinyl Rh-carbenoid intermediate, was developed. The results suggest that during its reaction with nitriles, the alpha-thiavinyl carbene acts as an umpolung 1,3-dipole equivalent, in contrast to its behavior during its reaction with alkynes. The developed method was successfully employed to synthesize pentaoligomeric arylene compounds consisting of three benzene and two isothiazole rings.

Tandem Photoarylation-Photoisomerization of Halothiazoles: Synthesis, Photophysical and Singlet Oxygen Activation Properties of Ethyl 2-Arylthiazole-5-carboxylates

The photochemical reaction between ethyl 2-chlorothiazole-5-carboxylate and benzene gave no product. In contrast, the reaction with ethyl 2-iodothiazole-5-carboxylate gave ethyl 3-phenylisothiazole-4-carboxylate. The same behaviour was observed if the reaction was performed in the presence of furan, thiophene and 2-bromothiophene. The products thus obtained were studied for their photophysical properties and it was found that the observed absorptions [lambda(max)=257 nm (epsilon = 7000 M-1 cm(-1)) for ethyl 3-phenylisothiazole-4-carboxylate, 278 nm (a = 7000 M-1 cm(-1)) for ethyl 3-(2-furyl)isothiazole-4-carboxylate, 269 and 285 nm (a = 7500 M-1 cm(-1)) for ethyl 3(2-thienyl)isothiazole-4-carboxylate] are mainly due to pi -> pi* transitions from the HOMO to the LUMO+1 orbital. The tested compounds showed fluorescence [lambda(em) = 350-400 (lambda(exc) = 300 nm), 360 and 410 nm (lambda(exc) = 320 nm), 412 nm (lambda(exc) 340 nm) for ethyl 3-phenylisothiazole-4-carboxylate, 397 and 460 nm (lambda(exc) = 300 nm), 381, 394 and 460 nm (lambda(exc) = 320 nm), 381, 398 and 466 rim (lambda(exc) = 340 nm) for ethyl 3-(2-furyl)isothiazole-4-carboxylate, 372, 377 and 414 nm (lambda(exc) = 300, 320 and 340 nm respectively) for ethyl 3-(2-thienyl)isothiazole-4carboxylate], possibly due to dual emission from different excited states. The use of the compound obtained as a sensitizer in the photo-oxidation of trans-alpha,alpha’-dimethylstilbene showed that all the new compounds are singlet-oxygen sensitizers.

Ruthenium(II) Complexes of Isothiazole Ligands: Crystal Structure, HSA/DNA Interactions, Cytotoxic Activity and Molecular Docking Simulations

Two new neutral ruthenium(II) complexes [Ru(eta(6)-p-cymene)Cl-2(1)] (3) and [Ru(eta(6)-p-cymene)Cl-2(2)] (4) (1=5-(phenylamino)-3-pyrrolidin-1-ylisothiazole-4-carbonitrile;2=3-morpholin-4-yl-5-(phenylamino)isothiazole-4-carbonitrile) have been synthesized and characterized using elemental analysis, IR, UV-Vis and NMR spectroscopy. The crystal structure was confirmed for complex3and both ligands. Examination of the interactions of ligands and complexes with CT-DNA (Calf Thymus DNA), as well as with HSA (Human Serum Albumin) revealed that ligands and complexes could interact with CT-DNA through intercalation and could bind strongly with HSA. Docking experiments toward DNA dodecamer indicate excellent accordance with experimental Delta G values. The cytotoxic activity of ligands and complexes was evaluated by MTT assay against HCT116 and HeLa tumoral cells. The complexes3and4showed good activity and selectivity on HCT116 cells. Neither of the tested compounds shows cytotoxic activity against a healthy MRC-5 cell line. Flow cytometry analysis showed the apoptotic death of the HCT116 cells with a cell cycle arrest in the S-phase.

Differences in kinetics of structurally related competitive GABA(A) receptor antagonists

Previously, 4-alkyl and 4-aryl substituted analogues of the low-efficacy partial GABA(A) receptor agonist 5-(4-piperidyl)-3-isothiazole (4-PIOL) have been identified as competitive GABAA receptor antagonists. These structurally related competitive antagonists show marked differences in their kinetic properties. The kinetics of 20 4-alkyl and 4-aryl substituted analogues of 4-PIOL, two 4-arylalkyl substituted 3-isothiazolol analogues and the classical GABAA receptor antagonist SR95531 was studied in cultured cerebral cortical neurons using whole-cell patch-clamp techniques. The kinetics of the antagonists was studied indirectly by measuring the changes in the response of the full GABAA receptor agonist isoguvacine (IGU) induced by concurrent application of an antagonist. When added, the majority of the antagonists did not affect the rate of deactivation of the IGU-induced responses. When removed, however, the majority of the antagonists slowed the reactivation phase of IGU implying that the dissociation of the antagonist from the GABAA receptor is the rate-limiting step. Surprisingly, the functional off-rates of the antagonists seemed to correlate better with the lipophilicity of the compounds than with the affinity and potency. This suggests that the dissociation of the tested antagonists from the GABAA receptor is restricted by lipophilic interactions, perhaps with the aromatic amino acids surrounding the GABA binding site. High-resolution infrared analysis of seven fundamental bands of gaseous isothiazole between 750 and 1500 cm(-1)

The gas phase IR spectrum of isothiazole, C3H3NS, between 550 and 1700 cm(-1) was recorded with a resolution of ca. 0.003 cm (-1). The rotational structure of seven fundamental bands in the region 750-1500 cm-1 has been assigned and analysed by the Watson Hamiltonian model. A number of local resonances in the bands have been identified and explained qualitatively in terms of Coriolis interactions. For each band upper state spectroscopic constants, including band center, rotational constants, and quartic centrifugal distortion constants are given. From observed crossings due to resonances we locate the weak bands v(9)(A’) and v(13)(A’) at 1041.9(2) and 642.0(3) cm(-1) respectively. The anharmonic frequencies have been determined using a cc-pVTZ basis set, at the MP2 and B3LYP levels; the two theoretical methods give very similar results for rotational constants, anharmonic band center frequencies and distortion constants, and many of these are in good agreement with experiment.
