Photooxidation of the Antimicrobial, Nonribosomal Peptide Bacitracin A by Singlet Oxygen under Environmentally Relevant Conditions

Bacitracin is a mixture of nonribosomal peptides (NRPs) that is extensively used as an antibiotic in both human and veterinary medicine. Despite its widespread use over the past six decades, very few studies have addressed the environmental fate of bacitracin and zinc-bacitracin complexes. In this study, the photochemical transformation of bacitracin components (i.e., cyclic dodecapeptides) in the aquatic environment was investigated. A high resolution mass spectrometry (HRMS)-based approach enabled monitoring of the photochemical degradation kinetics of individual bacitracin components, investigation of the relative contribution of reactive oxygen species (e.g., singlet oxygen, 1O2) in dissolved organic matter-sensitized photoreactions, and identification of oxidative modifications in bacitracin photoproducts. The results of this study support the hypothesis that indirect photochemical oxidation of the histidine (His) residue by 1O2 is a major degradation pathway for bacitracin A, the most potent congener of the mixture. Furthermore, the photooxidation rate of bacitracin A with 1O2 decreased upon bacitracin A coordination with Zn2+, demonstrating that the photochemistry of metal-bound His is different from that of metal-free His. Overall, these results provide insight into the fate of bacitracin components in the aquatic environment and highlight the potential of utilizing this HRMS-based methodology to study transformations of other environmentally relevant NRPs.

The crystal structures of [Tl(tsac)] (1) and [Tl(tsac)-(ophen)] (2) (tsac = anion of thiosaccharin; ophen = 1,10 phenanthroline) have been determined at 116 K by single crystal X-ray diffractometry. Complex 1 crystallizes in the monoclinic space group P21/a with Z = 4 and complex 2 in the monoclinic space group C2/c with Z = 8.In both complexes TI is coordinated to a thiosaccharinate anion through its sulphur and nitrogen atoms. A distorted eight fold coordination sphere around the cation in complex 1 is completed with two other longer Tl-S bonds and four Tl…O contacts with five symmetry related neighbouring thiosaccharinate anions. A phenanthroline molecule acting as a bidentate ligand through its nitrogen atoms completes a four-fold coordination around the metal atom in complex 2. The infrared spectra of both complexes were also recorded and their most important features discussed on the basis of its structural peculiarities.

Volatile composition and sensory and quality attributes of quince (Cydonia oblonga Mill.) fruits as affected by water stress

No information exists on the effect of water deficit on quince (Cydonia oblonga Mill.) fruit quality characteristics. In this paper, the effect of withholding irrigation water in the middle of the rapid fruit growth period on yield, main fruit physicochemical characteristics, sensory profile, and volatile composition of quince (cv. BA-29) was studied. The linear fruit growth phase was seen to be a critical phenological period for the marketable yield of quince because water deficit decreased the fruit weight, size, and moisture content. Also, this period was clearly critical for the fruit’s chemical characteristics. Water deficit induced important changes in an important number of the identified volatile compounds, characterized by decreases on pear, pineapple and apple sensory descriptors and increases on the green-herbaceous sensory descriptors. Moreover, total soluble solids increased and the total polyphenols content and antioxidant activity decreased as a result of water deficit effect, even though the still high levels of total polyphenols, regardless of the fruit water status, suggested that quinces can be used as an important source of natural antioxidants. The fruit sensory analysis indicated that the peel colour intensity of fruits from the water deficit treatment increased and fruit flesh texture improved. Indeed, the main handicaps for its consumption as fresh fruit (corkiness and fibreness) severely decreased and crunchiness increased.
