A mixture of 10 mmol of compound (IVa) as hydrochloride and 10 mmol of compound (IVb), 30 mmol of potassium carbonate, small crystal of potassium iodide and 20 ml of Nu,Nu-dimethylformamide was stirred at room temperature until disappearance of starting materials (TLC monitoring). Usually, the reaction was carried out for 2 days. The reaction mixture was subsequently poured into 50 ml of water, and precipitate thus formed was isolated by filtration. For purification, crude product was suspended in 20 ml of methanol, then the solid was filtered off and dried to constant weight. Alternatively (for 111-8), the reaction was carried out in acetonitrile, after completion of the reaction the solvent was evaporated and product was purified by column chromatography on silica gel using chloroform/methanol 95:5 as eluent.Structures of products were confirmed by mass spectrometry.According to the above procedure the following compounds were prepared:2-(4-(4-(1 ,2-benzothiazol-3-yl)piperazin-1 -yl)butyl)-1 H-isoindole-1 ,3(2H)-dione (111- 1 ), reaction in Nu,Nu-dimethylformamide, MS: 421 [M+H+], yield: 87%;

General procedure: The target compounds were synthesized using the procedure given in literature by refluxing aryl/heteroaryl acids with carbohydrazide 4a or 4b in phosphorous oxychloride. The reaction was monitored with the help of TLC to check its completion. After completion of reaction, the mixture was poured over crushed ice and allowed to precipitate. The precipitate was vacuum filtered, dried and recrystallized from methanol. Column chromatography was done on using n-hexane: ethylacetate mixture (7:3). Solvent was evaporated invacuo using rotary evaporator to obtain pure product.

To a suspension of isothiazole-3-carboxylic acid (2 g, 15.49 mmol) in DCM at 0 C was added oxalyl dichloride (1.3 equiv.) followed by three drops of DMF. Bubbling commenced, and the reaction was warmed to 23 C after 10 min. The mixture was stirred for 3 h, then cooled to 0 C. N,0-dimethylhydroxylamine hydrochloride (1.3 equiv.) was added to the reaction, followed by triethylamine (3.5 equiv.) which was added dropwise via syringe over 10 min. Reactions was warm slowly to 23 C overnight, and stirred for a total of 15 h. Reaction mixture was diluted with IN HC1 solution and dichloromethane (1 :1 ratio). Layers were separated, and the aqueous layer was extracted with DCM (2x). Combined organic layers were dried over magnesium sulfate, filtered, and the solvent was removed in vacuo. Crude reside purified via silica gel chromatography (utilizing a hexane/ethyl acetate mix as eluent) to deliver the desired intermediate, N-methoxy-N-methylisothiazole-3-carboxamide (1 g, 38%> yield) as a pale yellow solid. NMR (400 MHz, CDCI3) delta ppm 8.67 (d, 1 H), 7.69 (br s, 1 H), 3.80 (s, 3 H), 3.46 (br s, 3 H).

The crude product was eluted through a flash column (silica gel 60, 230-400 mesh, EtOAc) to give a clear, viscous oil which crystallized on standing. Yield = 1.16 g (42%); MS(APCI), (M + 1)+ = 433. 1H-NMR (DMSO-d6, delta) 8.03 (d, 2H, J = 9.0 Hz), 7.53 (t, 1H, J =8.1, 7.8 Hz), 7.40 (t, 1H, J = 8.1, 7.3 Hz), 6.87 (d, 2H, J = 7.3 Hz), 3.69 (t, 2H, J = 5.9, 5.9 Hz), 3.26-3.43 (m, 6H), 2.46-2.78 (m, 12H), 1.78 (m, 2H). EXAMPLE 1 3-(1-Piperazinyl)-1,2-benzisothiazole STR26 A mixture of 3-chloro-1,2-benzisothiazole (37.8 g., 0.235 mole) and piperazine (304.2 g., 3.53 mole) is heated under an argon atmosphere for a period of 20 hr. at 120¡ã C. in a closed reactor. The reaction mixture is dissolved in 2 liters of water and the aqueous solution repeatedly extracted with methylene chloride. Extracts are combined, dried over magnesium sulfate and concentrated in vacuo. Residual material is taken up in ether, filtered and concentrated in vacuo to afford 24.4 g. (47percent) of 3-(1-piperazinyl)-1,2-benzisothiazole free base as a viscous oil. A sample of the free base converted to the hydrochloride salt in ether with ethanolic hydrogen chloride and crytallized from methanol-ethanol affords analytically pure 3-(1-piperazinyl)-1,2-benzisothiazole hydrochloride, m.p. 280¡ã C. (dec.). Anal. Calcd. for C11 H13 N3 S.HCl: C, 51.66; H, 5.52; N, 16.43. Found: C, 51.34; H, 5.46; N, 16.16.

Step 2.To a solution of 1 (97.0 mg, 0.57 mmol) in DMF (4 mL) was added NaH (13.9 mg, 0.58 mmol) at 0¡ãC. The mixture was stirred at 0¡ãC for 30 minutes. Dichloropyrimidine 2 (100 mg, 0.48 mmol, 1.0 eq) was added, the solution was allowed to warm to 15¡ãC and stirred for 15 hours. The resulting mixture was poured into ice water (w/w = 1/1, 20 mL) and stirred for 10 minutes. The aqueous phase was extracted with EtOAc (30 mL) and the combined organic phase was washed with brine (20 mL), dried with anhydrous Na2S04, filtered, and concentrated in vacuo to afford 3 (120 mg, 72percent).

730mg of 3,4-dichloroisothiazole-5-carboxylic acid (3.7 mmol) were dissolved in 10 ml of dichloromethane and a drop of dimethylformamide was added. 1.4 g of oxalyl chloride (11.1 mmol) were added dropwise at room temperature. After stirring for 1 h at room temperature, the solution was evaporated to dryness on a rotary evaporator. The residue was taken up in 3 ml ofdichloromethane and slowly added dropwise to a solution of 626 mg of 1- cyclohexylmethanamine (5.5 mmol) and 746 mg of triethylamine (7.4 mmol) in 10 ml of dichloromethane. The mixture was stirred at room temperature for 1 h. The reaction mixture was then added to water and extracted repeatedly with dichloromethane. The concentrated extracts were dried over MgSO4, concentrated and purified by column chromatography. Yield:1.05 g (97percent of theory).?H-NMR (400 MHz, CDC13 , ppm) 6.86 (br, 1H), 3.34 (tr, 2H), 1.77 (m, 4H), 1.66 (m, 1H), 1.58 (m, 1H), 1.3-1.15 (m, 3H), 1.0 (m, 2H).
