A solution of compound 117 (0.35 g, 2.67 mmol), diphenylphosphoryl azide (0.57 mL, 2.67 mmol) and triethylamine (0.37 mL, 2.67 mmol) in tert-butanol (10 mL) was heated at reflux for 5 hours, at which time thin layer chromatography (DCM/Hexanes) indicates the reaction is complete. The reaction mixture was cooled to room temperature, poured into water and extracted with diethyl ether (3¡Á). The solid was collected by suction filtration, washing and triturating with H2O, then dried in a vacuum oven at 50C for 3 d to give the title compound (2.68 g, 72%) as a light brown amorphous solid. Piperazine (508.8 g) and Tert. Butanol (200 ml) were placed in round bottom flask (RBF) and then the resulting solution heated up to 90 to 100 degree C. 3-chloro-1,2-benzisothiozole (40 g) was added to the solution 5 times with in time interval of 5 to 20 minutes at the same temperature. Temperature was raised up to 110 to 130 degree C. and maintained for 16 hours. After confirmation of the completion of the reaction by TLS (Thin Layer Chromatography), the resulting solution was cooled to 80 to 90 degree C. followed by addition of water (800 ml). The above mixture was cooled to 25 to 35 degree C. and was filtered to remove the solid particles. Water (100 ml) was added to the filtrate and then pH was raised to 12-14 with caustic lye (75 ml). Toluene (400 ml) was added to the alkaline solution the resulting bi-phasic mixture was stirred vigorously for 15 to 30 minutes at 25 to 35 degree C. Organic layer separated and the aqueous layer was multiply extracted with toluene followed by combing all organic layers. The combined organic layer was washed with water (200 ml). Active carbon (10 g) was added to the washed organic layer and then filtered out the carbon. The organic solvent was evaporated till the volume reaches to 150 to 200 ml. The resulting concentrated solution was cooled to 0 to 5 degree C. and maintained for about 2 hours. Separated solids were filtered and then washed with chilled toluene (20 ml). Finally, Isolated compound was dried under reduced pressure to get titled compound 130 to 135 g.

The reaction was heated to 95 C. with stirring for 2 hours, cooled, and the dioxane evaporated. The resulting solution was diluted with water and brine then extracted four times with dichloromethane. The organic solution was dried over anhydrous sodium sulfate, filtered, and volatiles evaporated. The resulting material was purified using chromatography on a normal phase silica gel column with 0 to 10% ethyl acetate in hexanes. Fractions containing clean product were combined and the solvent evaporated. The material was dried under vacuum at room temperature to give the title compound (118 mg, 77%).

Excess dried, -325 mesh potassium carbonate (20 g) was diluted in 500 mL acetone and 3-piperazin-1-yl-benzoisothiazole hydrochloride (13.37 g, 52.4 mmol was added. The mixture was stirred for 15 min before 2-nitrophenethyl tosylate (15.3 g, 47.7 mmol) and catalytic 18-crown-6 (0.5 g, 1.9 mmol) was added. The mixture was stirred at reflux for 42 h. After cooling, the salts were filtered off and washed with acetone and the filtrate was concentrated. The residue was taken up in methylene chloride and washed with water. The organic layer was dried over sodium sulfate, and concentrated. The residue was triturated with ethyl acetate and the collected solid was washed with ethyl ether and dried in vacuo to afford 13.08 g of a viscous, brown liquid. Yield 75%; MS (APCI): 369 [M+H]+.

The product was recrystallized from n-hexane/CH2Cl2 at 4C and furnished orange crystals of analytically pure 1 in 12% yield (14mg). Data for 1: Anal. Calc. for C20H8N2O10Ru2S4: C, 31.33; H, 1.05; N, 3.65. Found: C, 31.64; H, 1.12; N, 3.69%.
